Abstract:
A method for discrimination between Korean beef cattle and imported cattle using a multiplexing PCR and a primer therefore are provided to distinguish species of cattle by analyzing gene loci rapidly and accurately. A method for discrimination between Korean beef cattle and imported cattle using a multiplexing PCR comprises the following steps of: obtaining DNA of cattle having two pairs of primers and a plurality of gene loci; amplifying a base sequence comprising each locus of a DNA sample into a multiplexing PCR; and measuring characteristic form and quality of an allele of each microsatellite locus.
Abstract:
A process for genotyping the cSNPs(complementary single nucleotide polymorphism) in CFB(complement factor B) gene localized to the SLA(swine leukocyte antigen) class III is provided to improve rapidness and economical efficiency of the genotyping as compared with conventional PCR-RFLP (polymerase chain reaction- restriction flagment-lengh polymorphism) by simultaneously analyzing cSNPs with multiplex PCR. A process for simultaneously genotyping three cSNPs in CFB gene localized to the SLA class III comprises the steps of: (1) preparing 3 fluorescence-labeled primer sets containing primers having the nucleotide sequences of SEQ ID NO:1 to SEQ ID NO:9 by using ARMS(amplification refractory mutation system); (2) performing multiplex PCR of a DNA sample of pig by using the primer sets; and (3) subjecting the PCR products to electrophoresis and analyzing the genotypes.
