Abstract:
본 발명은 셀로비오스 디하이드로게나제를 공유결합(covalent binding)을 이용하여 담체에 고정화시키기 전에 전처리하는 방법에 관한 것이다. 구체적으로 셀로비오스 디하이드로게나제를 담체에 고정화시키기 전에 셀로비오스 디하이드로게나제 용액에 단당류, 이당류, 다당류 또는 이들의 혼합물을 첨가하여 반응시키는 전처리 과정을 통하여 공유결합에 의한 셀로비오스 디하이드로게나제의 고정화 시 발생하는 활성도의 감소를 억제하고 여러 번 재사용할 경우에도 셀로비오스 디하이드로게나제의 활성을 유지할 수 있다.

PURPOSE: A mannanase isolated from Clostridium cellulovorans is provided to ensure excellent enzyme activation and to effectively produce biofuel from ligneous biomass. CONSTITUTION: A mannanase isolated from Clostridium cellulovorans contains has an amino acid sequence of sequence number 1. A gene encoding the mannanase has a base of sequence number 2. A recombinant vector contains the gene. A method for decomposing ligneous biomass comprises a step of treating the ligneous biomass with the mannanase. A composition for decomposing ligneous biomass contains a complex of mini CbpA and endoglucanase E as an active ingredient.

PURPOSE: A yeast transformant containing a chimera endoglucanase gene and small cellulose-binding protein gene is provided to develop feed and probiotics. CONSTITUTION: A recombinant vector, pADHcCelEmCbpA contains: a chimera gene in which dockerin module is linked to a Clostridium sp.-derived endoglucanase active site gene; and mCbpA gene which is a part of cellulose-binding protein-A derived from Clostridium sp. The endoglucanase active site contains an amino acid sequence of sequence number 1.

PURPOSE: A cohesin bio-marker and a method for detecting cellulosome forming enzyme using the same are provided to develop an artificial enzyme complex system. CONSTITUTION: A marker composition for detecting cellulosome forming enzyme contains cohesin domain as an active ingredient. The cohesin domain is labeled as a marker of fluorescence or radioactive isotope. A method for detecting cellulosome forming enzyme comprises: a step of culturing microorganism which decomposes cellulose to obtain proteins; a step of isolating the proteins; a step of treating a marker composition containing cohesin domain as an active ingredient; and a step of confirming complex of the marker and protein.
