An applicator A is configured to transfer an active ingredient into a body through skin by a puncture in the skin with microneedles 32. The applicator A includes a piston plate 20 that is provided with a protrusion part 33 and protrusion parts 20d 1 to 20d 4 on a lower surface side of a main body 20a of the piston plate. The microneedles 32 are provided on a surface of the protrusion part 33. The protrusion parts 20d 1 to 20d 4 are provided in a periphery of the protrusion part 33 while spaced apart from the protrusion part 33.

The present invention addresses the problem of providing a low-molecular weight adjuvant which can be administered safely without inducing skin irritation or the like by transdermal or transmucosal administration and is for efficiently enhancing the immunogenicity of an antigen. The present invention relates to an adjuvant for transdermal or transmucosal administration, containing at least one member selected from polyhydric alcohols and derivatives thereof including glycerol, glycerol derivatives and the like.

A puncture device which can be made smaller in size and lighter in weight and a method for manufacturing the same are provided. A puncture device 1 is a device for transferring an active ingredient into a body through a skin. The puncture device 1 comprises a tubular housing H, a piston P slidably arranged within the housing H, and a conical coil spring 40 arranged within the housing H and providing the piston P with a biasing force. The piston P has a piston body 20 and a microneedle member 30 engaging the piston body 20 so as to integrate therewith. The microneedle member 30 has a base 31 and a plurality of microneedles projecting from a surface of the base 31.

An applicator comprises a transmission member, a housing guiding a reciprocation of the transmission member, and a cap covering the housing. The transmission member is provided with a stopper extending in a direction orthogonal to an axial direction thereof. An upper end of the housing is formed with a projection sliding the stopper. The inside of the cap is formed with a lead part. The stopper is slid by the lead part to an end part of the projection when the cap is pushed and thereafter disengages from the projection when the cap is further pushed, so that the transmission member transmits the biasing force of an elastic member to a microneedle. This enables the microneedle to be applied to a skin.

It is intended to provide a low molecular weight adjuvant which can be administered safely without inducing skin irritation or the like by transdermal or transmucosal administration and is for enhancing immunogenicity of an antigen efficiently. It can be achieved by an immunostimulant adjuvant containing at least one substance selected from the group consisting of aliphatic alcohols, free fatty acids and fatty acid derivatives but does not contain a substance represented by the following formula. (In the formula, R 3 and R 4 may be combined to form a cyclic ring, and R 1 and R 2 independently represent an alkyl side chain having 1 to 16 carbon atoms.)
