Abstract:
A skin external composition improving anti-aging through the regulation of caveolin productivity is provided to promote the collagen biosynthesis, prevent skin aging and enhance skin elasticity. A skin external composition comprises 0.001-10% by weight of caveolin or a material which increases the caveolin biosynthesis as a efficient ingredient. The material which increases the caveolin biosynthesis is the cholesterol. The skin external composition is used in a form of skin, lotion, massage cream, pack, gel, ointment, gel, cream, patch, or transdermal administration of spray. A screening process is performed by selecting materials which increase the caveolin productivity in dermis from candidate materials in vitro.

A method for diagnosing degree of aging and a kit thereof are provided to rapidly diagnose degree of aging caused by female hormone deficiency using a marker gene and induce development of a skin aging inhibitor. A kit for diagnosing degree of aging caused by female hormone deficiency comprises one or more polynucleotides and polynucleotide containing ten or more continuous nucleotide as the fraction of the polynucleotides or complementary polynucleotide thereof. The kit for diagnosing degree of aging caused by female hormone deficiency contains a solid support whose surface is coupled with the polynucleotides. If the quantify of transcriptome mixed to the polynucleotides among transcriptomes of keratinocyte is less than standard value, the skin is diagnosed as geroderma.

A cosmetic composition is provided to prevent the damage of hair by reducing the loss of labile proteins inside hair. A cosmetic composition for preventing the damage of hair contains 0.01-10 wt% of a lime oil, a lemon oil, or their mixture. Preferably the lime oil is an oil extracted by distilling Citrus aurantifolia, and the lemon oil is an oil extracted by compressing the shell of Citrus limon.

A skin external composition comprising a cathepsin G inhibitor is provided to prevent an adhesion protein from being degraded by intrinsic aging and photo-aging, reduce the production of metalloproteinase and inhibit the activity of metalloproteinase, thereby being more potent anti-aging substance than existing anti-aging substances. A method for screening anti-aging materials is provided to develop the anti-aging materials which effectively inhibit the quantitative and qualitative increase of metalloproteinase, resulting from the degradation of an adhesion protein by cathepsin G in skin cells without direct UV irradiation. A skin external composition for preventing skin aging comprises 0.01-10 wt% a cathepsin G inhibitor as an effective ingredient, wherein the cathepsin G inhibitor is at least one selected from the group consisting of compounds represented by formulae(1) to (4). The composition is formulated into skin lotion, astringent lotion, milk lotion, nourishing cream, essence, massage cream, eye cream, pack, body lotion, body cream, body oil or body essence. A method for screening an anti-aging material comprises the steps of: (a) adding an anti-aging candidate material to a substrate in the presence of cathepsin G enzyme; and (b) measuring the inhibition of activity of the cathepsin G enzyme. Further, the measure of the inhibition of activity of the cathepsin G enzyme is performed by measuring a concentration of a produced metalloprotease.

An anti-aging composition is provided to promote activity of proteasome of fibroblast and keratinocyte, thereby inhibiting accumulation of oxidation proteins and blocking increase of aging markers. An anti-aging cosmetic or pharmaceutical composition comprises 0.001-5 wt.% of at least one selected from the group consisting of epicatechins, caffeine, naringenin, ellagic acid, sanguinarine, eupomatenoid 6, mahanimbine, tanshinone IIA, theobromine, and gallic acid as an effective ingredient. Further, a 0.001 to 5 wt.% of effective ingredient is comprised based on the 100 wt.% of composition.

A composition comprising minoxidil, tretinoin(all-trans retinoic acid), a solvent and glycerine is provided to promote permeation of the minoxidil without stimulus of the tretinoin, thereby inducing hair growth and alleviating alopecia. An external composition for preventing hair loss and promoting hair growth comprises 0.5-7.5 weight/volume% of minoxidil as a hair growth ingredient, 0.007-0.015 weight/volume% of tretinoin, 72-83 weight/volume% of ethanol, and 15-20 weight/volume% of glycerine.

A screening method of pigmentation regulatory agents by using zebrafish(Danio rerio) is provided to screen a large quantity of materials capable of increasing or inhibiting melanin formation as pigmentation regulatory agents easily and rapidly by determining various biological parameters from zebrafish. A screening method of pigmentation regulatory agents by using zebrafish comprises the steps of: administering a candidate pigmentation regulatory agent into an embryo of zebrafish; and detecting the reactions of zebrafish including expression amount of melanin formation, and in vivo activity, expression amount or transition amount of proteins associated with melanin formation, wherein the screening method optionally comprises a detecting step of toxicity of the candidate material by measuring morphological change or heartbeat number.

A skin external composition for skin-whitening containing manassantin B is provided to inhibit mobility of melanin by inhibiting binding of melanophilin as a carrier of melanosome in a melanocyte with myosin 5a, thereby forming melanosome aggregation and whitening the skin. A skin external composition for skin-whitening contains 0.001-10% of manassantin B which is extracted from Saururus chinensis Baill by extracting Saururus chinensis Baill with water or organic solvent selected from ethanol, methanol, butanol, ether, ethylacetate, chloroform and methylene chloride, filtering the extract, concentrating the filtered solution under reduced pressure, extracting the concentrate with methylene chloride and concentrating the extract under reduced pressure, and is formulated as solution, suspension, emulsion, paste, gel, cream, lotion, powder, soap, surfactant-containing cleansing, oil, powder foundation, emulsion foundation, wax foundation or spray.
