Abstract:
본 발명은 TGase2(Transglutaminse 2)가 VHL(Von Hippel-Lindau)을 조절하여 암형성과정 중 HIF-1α(Hypoxia-inducible factor-1α), IGF-1α(Insulin-like growth factor receptor) 및 VEGF(Vascular endothelial growth factor)의 안정성 및 NF-κB(Nuclear factor kappa-light-chain-enhancer of activated B cells)의 활성을 조절한다는 것을 규명하고, VHL 단백질 수준 또는 VHL의 중합을 측정함으로서 TGase2 억제제 또는 촉진제를 검출하는 방법을 제공한다. 또한 HIF-1α, IGF-1R 또는 VEGF의 단백질 수준을 추가로 측정함으로써 TGase2 억제제 또는 촉진제를 검출하는 방법을 제공한다. 본 발명에서 TGase2 억제제는 항암제로서, 또한 TGase2 촉진제는 바이러스 감염 질환 치료제로서 유용하게 사용될 수 있다.

PURPOSE: A novel pyrazolodiazepine compound is provided to strongly suppress transglutaminase and to prevent and treat diseases caused by transglutaminase over-activation. CONSTITUTION: A pyrazolodiazepine compounds have a structure of formula I. A pyrazolodiazepine-8(2H) thion compounds has a structure of formula Ia. A pyrazolodiazepine-8-yl thio compounds has a structure of formula Ib. A suppressor to transglutaminase contains the compound of formula I or pharmacologically acceptable salt thereof as an active ingredient. A pharmaceutical composition for preventing and treating diseases caused by transglutaminase contains the compound of formula I or pharmacologically acceptable salt as an active ingredient. The diseases caused by transglutaminase are neural diseases or cancer.

A cancer sensitizer is provided to make cancer cells highly sensitive to anticancer agents without side effects in contrast to conventional chemical sensitizers by comprising glucosamine which is a natural material, or glucosamine derivatives. A cancer sensitizer comprises glucosamine, a glucosamine derivative or a salt thereof, wherein the glucosamine derivative is represented by the formula(2). In the formula(2), R is an acyl group having 2 to 18 carbon atoms or a straight or branched alkyl group having 1 to 5 carbon atoms. A composition for inhibiting chemoresistance comprises glucosamine, a glucosamine derivative, or a salt thereof, and a pharmaceutically acceptable carrier. An anticancer composition comprises the composition for inhibiting chemoresistance and an anticancer agent selected from the group consisting of mechlorethamine, chlorambucil, phenylalanine, mustard, cyclophosphamide, ifosfamide, carmustine(BCNU), lomustine(CCNU), streptozotocin, busulfan, thiotepa, cisplatin, carboplatin, dactinomycin(actinomycin D), doxorubicin(adriamycin), daunorubicin, idarubicin, mitoxantrone, plicamycin, mitomycin, C bleomycin, vincristine, vinblastine, paclitaxel, docetaxel, etoposide, teniposide, topotecan and irinotecan. A method for detecting an inhibitor or a promoter of transglutaminase 2(TGase 2) is provided to effectively screen the inhibitor or promoter of the TGase2 by measuring the level of I- kappaBalpha protein, the polymerization of I-kappaBalpha protein and the activity of NF-kappaB. The method comprises the steps of: (a) treating cell expressing I-kappaBalpha and NF-kappaB with a candidate inhibitor or a activator; (b) inducing the expression of TGase 2 using lipopolysaccharide(LPS), UV, glutamate, calcium inophore, maitotoxin, retinoic acid(RA), inflammation-inducing cytokine, oxidative environment or virus inflammation; and (c) identifying the level of the I-kappaBalpha protein, the polymerization of the I-kappaBalpha protein and the activity of NF-kappaB with a control.

An apparatus and a method for detecting approaching vehicles and an outside mirror using the same are provided to prevent car accidents entering dead spaces by extracting motion vector from image obtained by photographing the rear part of the vehicle. An apparatus and a method for detecting approaching vehicles and an outside mirror using the same comprise a camera(110), and a controller(120). The camera photographs the rear area of a vehicle. The controller sets up one image as the standard image among a plurality of consecutive images which the camera photographs. The motion vector of the pixels in the standard image is detected with reference to one or more images among before or after images of the standard image. In case the traveling direction of the detected motion vector is analyzed and the pixels having the motion vector in the approaching direction is detected, the controller outputs alarm signal according to the access of objects.
