Abstract: 기존에 FXIII의활성도는주로 ELISA를통하여이루어져왔다. 그러나이와같은방법은활성도측정에시간이많이소요되고, 다량의시료를필요로하고, 재현성이떨어진다. 따라서본 발명에서는 FXIII의기질중 하나인피브리노겐을어레이형태로제작하여 FXIII의아미드전이반응(transamidation)을유도시킴으로써감도가높은형광으로표지된검출자를이용해형광값을측정하여세포나혈액내의 FXIII의양을측정한다. 따라서본 발명은극히소량의시료만으로쉽고빠르게분석가능하며, 각종질환검사에활용할수 있다.

Abstract: PURPOSE: A method for measuring activity of blood coagulation factor XIII is provided to quickly and easily analyze activity of blood coagulation factor XIII and to use in various diseases tests. CONSTITUTION: A method for measuring activity of blood coagulation factor XIII comprises: a step of preparing an array chip in which fibrinogen is fixed on a plurality of spots; a step of dropping reagent containing thrombin, Ca^2+, and 5-(biotinamido)pentylamine with a sample containing blood coagulation factor XIII on a fibrinogen to induce transamidation; a step of introducing an analysis marker to the biotin; and a step of measuring activity level of the blood coagulation factor XIII.
Abstract translation: 目的:提供一种测定血液凝固因子XIII活性的方法,以快速,方便地分析凝血因子XIII的活性,并用于各种疾病检测。 构成:测定凝血因子XIII的活性的方法,其特征在于,包括以下步骤:制备阵列芯片,其中纤维蛋白原固定在多个斑点上; 将含有凝血酶,Ca 2+和5-(生物素酰氨基)戊胺的试剂与含有凝血因子XIII的样品滴加在纤维蛋白原上以引发转酰胺化步骤; 将分析标记物引入生物素的步骤; 以及测定凝血因子XIII的活性水平的步骤。
