Abstract: 본 발명은 글루타티온 S 트랜스퍼라제 태깅(GST tagging) 단백질 분리용 나노입자와 제조 방법 및 이를 이용한 단백질 분리 정제 방법에 관한 것으로서, 보다 상세하게는 표면에 글루타티온기가 결합된 자성 나노입자를 이용하여 글루타티온 S 트랜스퍼라제 태깅(GST tagging) 단백질을 분리 정제하는 방법에 관한 것이다. 본 발명에 따르면 특정 단백질을 매우 선택적으로 간단하고 신속하게 분리할 수 있어, 암과 같은 질병 치료 연구에 유용하게 사용될 수 있다.

Abstract: PURPOSE: A nanoparticle for isolating proteins is provided to isolate and purify GST-tagged protein using a magnetic nanoparticle on which glutathione is conjugated. CONSTITUTION: A nanoparticle for isolating GST-tagged protein has a magnetic nanoparticle surface on which glutathione group is conjugated. The magnetic nanoparticle is conjugated with a silica layer. The glutathione group is conjugated with 2-pyridyl disulfide which is bound to the silica layer. A method for manufacturing the nanoparticle for isolating the GST tagged protein comprises: a step of coating the surface of magnetic nanoparticle with the silica layer; a step of binding 2-pyridyl disulfide group to the silica layer; and a step of binding glutathione group to 2-pyridyldisulfide group.

Abstract: A nanoparticle for peptide separation and a preparation method thereof are provided to rapidly and selectively separate desired peptide through a simple process owing to magnetic nanoparticles of which the surface is coupled with a first functional group which selectively reacts to a certain peptide. A nanoparticle for peptide separation has the magnetic surface which is coupled with a first functional group having thiol selectivity. The nanoparticle for peptide separation selectively couples with the peptide including cysteine. The first functional group includes 2-pyridyl disulfide residual group. The magnetic nanoparticle is coated with a silica layer. The first functional group is bonded with the silica layer. The surface of the nanoparticle additionally contains a second functional group which prevents the coherence of the nanoparticles.
