Abstract:
PURPOSE: A plant transformation vector, pCMF, for removing a selective marker is provided to enable easy modification of the vector and transduction of a target gene, and to develop marker-free GM(genetically modified) crops. CONSTITUTION: A site-specific recombinase gene, mFLP, has a base in sequence number 1. A vector for transformation in a plant contains the gene, mFLP. The vector contains a stress-induced promoter and recombinase recognition site. The vector is able to remove a selective marker by induction of mFP expression and self-cleavage due to stress. The vector contains MCS(multi-cloning site). A plant is transformed with the vector. Agrobacterium tumefaciens LBA4404 is transformed by the vector and is used as a plant carrier. A transgenic plant is prepared by transforming with the vector and is able to remove the selective marker.
Abstract:
PURPOSE: A method for simply detecting genetically modified corns is provided to shorten time for analyzing PCR result without specialized knowledge or equipment. CONSTITUTION: A method for detecting genetically modified corns comprises: a step of preparing primer sets which are each complementary to CaMV 35S promoter and SSIIb(starch synthase IIb); a step of performing PCR using the primer sets; a step of flowing PCR products on a strip on which an anti-secondary labeling substance antibody and an anti-tertiary labeling substance antibody are separately attached; and a step of determining corns and genetically modified corns by luminescence of the anti-tertiary labeling substance antibody and anti-secondary labeling substance antibody, respectively.

본 발명은 식물 선발마커 제거 식물 형질전환체의 제조를 위한 벡터와 이를 이용한 식물 형질전환체의 제조방법에 관한 것으로서, 그 구성은 마커-프리(marker free) 형질전환 식물체 제조를 위해 스트레스 유도성 프로모터에 의한 부위 특이적 재조합 유전자의 발현을 이용하여 선발마커를 제거할 수 있는 벡터를 제조하고, 상기 벡터를 이용하여 형질전환 과정에서 발생되는 스트레스에 의해 자발적으로 식물선발마커 유전자가 제거된 형질전환체를 제조하는 과정과, 선발표지를 이용하여 획득한 형질전환체를 재분화를 통해 식물선발마커를 제거하는 제조 방법을 제공한다. 이러한 방법은 식물 형질전환체내에서 식물선발마커를 제거하기 위해 사용되는 동시형질전환방법에 비해 형질전환 효율이 높고 선발표지 제거 형질전환체의 제조가 간편하며 자가 및 타가수정식물의 적용이 가능하다. 스트레스 유도성 프로모터, 선발마커 제거 발현 벡터, 선발마커 제거 식물체,
