Abstract:
본 발명은 단일사슬항체조각(scFv)과 페리틴이 결합된 형태의 융합단백질, 상기 융합단백질을 코딩하는 폴리뉴클레오티드, 상기 폴리뉴클레오티드를 포함하는 발현벡터, 상기 발현벡터가 도입된 형질전환체, 상기 형질전환체를 배양하고 이로부터 상기 융합단백질을 제조하는 방법, 상기 융합단백질을 포함하는 나노클러스터, 상기 나노클러스터를 포함하는 약물 전달체, 진단키트, 단백질 칩 및 상기 나노클러스터를 이용하는 항원 검출방법에 관한 것이다. 본 발명은 단일사슬항체조각(single chain variable fragment;, scFv) 의 서열을 페리틴에 도입함으로써 융합단백질을 제조하고 이를 이용하여 항체의 항원인식부위 24개가 항체의 페리틴표면에 발현되게 함으로써 바이오칩, 약물전달시스템, 질병진단 및 치료제 개발 등에 활용할 수 있다.

PURPOSE: A natural food preservative is provided to minimize the generation of problems caused by the toxicity and irritation effect of a chlorine based chemical preservative. CONSTITUTION: A natural food preservative contains 1-2 parts of paeonia lactiflora extract by weight, 1-2 parts of cinnamomum loureirii extract by weight, and 3-8 parts of rhus japonica extract by weight. The extracts are obtained by the following steps: heat-extracting paeonia lactiflora, cinnamomum loureirii, and rhus japonica extract at 50-90 deg C for 30-200 minutes after mixing with solvents, separately; decompress-concentrating each extract at 20-80 deg C for 60-200 minutes; and freeze-drying the obtained concentrate.

PURPOSE: A natural food antioxidant is provided to reduce the problems of the toxicity irritant property of a chemistry preserver for preserving fruits and vegetables. CONSTITUTION: Paeonia japonica, gallnuts, and Glycyrrhizae radix are separately mixed with solvents, and heated at 50-90°C for 30-200 minutes. Each extract is filtered, and decompress concentrated at 20-80°C for 60-200 minutes. Each concentrate is freeze-dried, and the concentrations of each concentrate become the same. 2-3 parts by weight of Paeonia japonica extract, 5-6 parts by weight of gallnut extract, and 1-2 parts by weight of Glycyrrhizae radix extract are mixed. PURPOSE: A natural food preservative containing a glycyrrhizae radix extract, a paeonia suffruticosa andrews extract, and a coptis japonica extract are provided to offer the excellent antibiotic and anticancer effect. CONSTITUTION: A producing method of a natural food preservative comprises the following steps: separately mixing glycyrrhizae radix, paeonia suffruticosa andrews, and coptis japonica with alcohol, and heating the mixture to extract; filtering the obtained extracts before decompress-concentrating; freeze-drying the obtained concentrates; and mixing the obtained concentrates in the same concentration.

PURPOSE: A method for gene overexpression is provided to promote expression of a target gene thereby producing a large amount of protein, and to regulate gene expression by a non-invasive method and specific mechanism. CONSTITUTION: A method for gene overexpression comprises: a step of transforming a mammalian cell by a vector containing a wave-sensitive promoter and a gene and preparing a transformed cell; and a step of culturing the transformed cells by 50 Hz of extremely low frequencies and 0.1mT of electromagnetic fields. The wave-sensitive promoter is human telomerase transcriptase. The gene is a heterogeneous gene. The mammalian cells are human kidney cell line, HEK293T. Proteins are prepared by an overexpression method. The protein is Staphylococcus SEB.
