Abstract: PURPOSE: A monoclonal antibody which specifically binds to undifferentiated human embryonic stem cells and a hybridoma producing the same are provided to study human embryonic stem cells. CONSTITUTION: A monoclonal antibody 297-D4 which is specific to undifferentiated human embryonic stem cells contain heavy chain variable domain(VH) with an amino acid sequence of sequence number 1 and a light chain variable domain(VL) with an amino acid sequence of sequence number 2. The monoclonal antibody is IgG1.k isotype and recognizes undifferentiated human embryonic stem cell BAP31-like proteins. A composition for removing the stem cells contains monoclonal antibody 297-D4. A kit for detecting the stem cells contains 297-D4. A method for isolating the stem cells comprises: a step of adding the monoclonal antibody 297-D4 to a cell group; and a step of isolating cells which combined with the monoclonal antibody 297-D4.
Abstract translation: 目的:提供特异性结合未分化的人胚胎干细胞的单克隆抗体和产生其的杂交瘤以研究人胚胎干细胞。 构成:对未分化的人胚胎干细胞特异性的单克隆抗体297-D4含有具有序列号1的氨基酸序列的重链可变区(VH)和具有序列氨基酸序列的轻链可变结构域(VL) 单克隆抗体是IgG1.k同种型并识别未分化的人胚胎干细胞BAP31样蛋白。 用于除去干细胞的组合物含有单克隆抗体297-D4。 用于检测干细胞的试剂盒含有297-D4。 分离干细胞的方法包括:将单克隆抗体297-D4加入细胞组的步骤; 以及分离与单克隆抗体297-D4组合的细胞的步骤。

Abstract: PURPOSE: A monoclonal antibody which specifically binds to undifferentiated human stem cells is provided to research difference of initial embryo development between rat and human. CONSTITUTION: An undifferentiated human embryonic stem cell-specific monoclonal antibody 247-B9 contains variable region of heavy chain having an amino acid of the sequence number 6 and variable region of light chain having an amino acid of the sequence number 8. The monoclonal antibody 247-B9 recognizes about 37kDa protein of undifferentiated human embryonic stem cell and hnRNP A2/B1 analog protein. A composition for removing the undifferentiated human embryonic stem cell contains the monoclonal antibody 247-B9.
