Abstract:
PURPOSE: A genetic map of Brassica rapa provided to improve the quality of Brassica rapa and to identify an useful gene which is used in protecting crops from diseases. CONSTITUTION: A kit for identifying SSR marker for constructing a genetic map of Brassica rapa contains a primer pair and reagent for PCR. The reagent contains thermoresistant DNA polymerase, dNTPs, and buffer. A method for identifying the SSR marker for constructing the genetic map for Brassica rapa comprises: a step of isolating genomic DNA from Brassica rapa; a step of amplifying a target sequence by PCR; and a step of detecting the PCR product. A method for selecting SSR primer set for constructing the genetic map comprises: a step of selecting BAC clone from base sequence of BAC clone; and a step of designing SSR primer set for 1-3 genetic maps per BAC clone from flaking base sequence of SSR motif.
