Abstract: PURPOSE: Provided is an expression vector for a mammal cell containing a short interpersed elements sequence (SINE) wherein SINE inserts an exogenous gene into chromosome, to enhance transformation rate. And a method for transformation using the same is also provided. CONSTITUTION: An expression vector used in a mammal cell for increasing transformation rate has more than one SINE at either 5'end or 3'end, or at both 5'end and 3'end of an exogenous gene. SINE can be B1 sequence, B2 sequence, fusion sequence thereof, ID sequence, Alu sequence or C-BCS sequence, and preferably fusion sequence of B1 and B2 sequence produced by typical ligation reaction. The vectors prepared by using PGKβ -geobp(A) as exogenous gene are as follows: (a) p(B1)2β-geo wherein 2B1 sequences locate at 5'end and 3'end of exogenous gene, respectively; (b) p(B2)2β-geo wherein 2B2 sequences locate at 5'end and 3'end of exogenous gene, respectively; (c) p(B1/2)β -geo wherein fusion sequence of B1 and B2 locates at 3'end of exogenous gene; (d) p(B1/2)2β-geo wherein fusion sequence of B1 and B2 sequences locates at 5'end of exogenous gene.

Abstract translation: 目的：提供含有短内插元件序列（SINE）的哺乳动物细胞的表达载体，其中SINE将外源基因插入到染色体中，以提高转化率。 还提供了使用该方法进行变换的方法。 构成：用于增加转化率的哺乳动物细胞中的表达载体在外源基因的5'端或3'末端或5'端和3'末端具有多于一个的SINE。 SINE可以是B1序列，B2序列，其融合序列，ID序列，Alu序列或C-BCS序列，优选通过典型连接反应产生的B1和B2序列的融合序列。 通过使用PGKβ-geobp（A）作为外源基因制备的载体如下：（a）p（B1）2β-geo，其中2B1序列分别位于外源基因的5'端和3'端; （b）p（B2）2β-geo，其中2B2序列分别位于外源基因的5'端和3'端; （c）其中B1和B2的融合序列位于外源基因的3'端的p（B1 / 2）β-geo; （d）p（B1 / 2）2β-geo，其中B1和B2序列的融合序列位于外源基因的5'端。
