Abstract: PURPOSE: A novel exochitinase isolated from Pseudoalteromonas issachenkonii is provided to ensure high activity at wide temperature and pH concentration range and to maintain enzyme activity. CONSTITUTION: An exochitinase has an amino acid sequence of sequence number 1. The exochitinase is derived from Pseudoalteromonas issachenkonii KOPRI 22718(KCTC 11415BP). An exochitinase has a nucleotide sequence of sequence number 2. A recombinant vector contains the exochitinase gene. A recombinant microorganism is obtained by inserting exochitinase gene to host cell chromosome or introducing the recombinant vector to a host cell. The exochitinase is obtained by culturing the recombinant microorganism.
Abstract translation: 目的:提供从假假单胞菌属假单胞菌分离得到的新型外切蛋白酶,以确保在宽温度和pH浓度范围内的高活性,并保持酶活性。 构成:外切蛋白酶具有序列号1的氨基酸序列。外切蛋白酶衍生自假金黄色葡萄球菌KOPRI 22718(KCTC 11415BP)。 外切蛋白酶具有序列号2的核苷酸序列。重组载体含有外切蛋白酶基因。 通过将外切蛋白酶基因插入宿主细胞染色体或将重组载体引入宿主细胞获得重组微生物。 通过培养重组微生物获得外切蛋白酶。

Abstract: A novel endochitinase and Sanguibacter sp. producing the same endochitinase are provided to show high activity of the endochitinase at various temperature including cool temperature and pH ranges, and stable activity in the presence of surfactants, so that the novel endochitinase and its producing microorganism are useful for production of chitinoligomer from chitin. A gene encoding the novel endochitinase has the nucleotide sequence of SEQ ID NO:1. The Sanguibacter sp. KCTC 10714BP produces the novel endochitinase encoded by the gene of SEQ ID NO:1. The novel endochitinase is produced by (a) culturing Sanguibacter sp. KCTC 10714BP or transformed Escherichia coli containing the endochitinase gene in a medium containing 0.1-1% of chitin as a carbon source at 4-25 deg.C, and (b) recovering the novel endochitinase from the cultured medium.
