    Abstract: A method of analysing a sample of multiple protein species comprising: separating proteins by species such that the multiple protein species emerge in a sequential order; digesting the separated proteins in said order; introducing the digested proteins into a mass spectrometer (MS) in said order wherein, within a time window, the digested proteins introduced into the MS are covariant. A portion of the separated protein may remain undigested and be introduced into the MS in conjunction with the corresponding covariant digested protein. A method of preparing a sample for use in matrix-assisted laser desorption ionization (MALDI) is also claimed. This may be carried out within a microfluidic device; said device having an inlet, means for separating proteins, an outlet connected to an inlet of a digestion chamber, the outlet of which is coupled to an interface to a MS, wherein said digestion chamber includes surfaces for immobilizing enzymes, and said device possesses an additional channel connecting the separation channel outlet directly to the MS interface.

    Abstract: The present invention provides, inter alia, an apparatus 1 for ionizing an analyte sample with a mass calibrant 5. The apparatus 1 includes an ionization chamber 10 defining an ionization region. A first passageway 12 is coupled to the ionization region for delivering the analyte sample to the ionization region and a second passageway 20 leading to a mass analyzer having an orifice 22 is arranged adjacent to the ionization region to receive ions from the ionization region. Finally a third passageway 30 is coupled to the ionization chamber 10 at a first end and has a second end with an orifice arranged to receive gaseous neutral mass calibrant molecules. An ionization device 15 arranged within the ionization chamber 10. In addition, the present invention also provides calibrant formulations that release mass calibrants in a slow, controlled manner wherein the mass calibrant is combined with a moderator substance which due to its lower vapor pressure with respect to the mass calibrant itself allows a slow controlled evaporation rate.

The apparatus 1 includes an ionization means 15 within an ionization chamber 10, a first passageway 12 for delivering the analyte sample to the ionization chamber, a second passageway 20 leading to a mass analyzer, and a third passageway 30 coupled to the ionization chamber for delivering a mass calibrant 5. Primary ions of either the analyte sample or mass calibrant are generated within the ionization chamber, and gaseous neutral molecules of either the mass calibrant or analyte sample respectively are ionized through interaction with said primary ions. In addition, the present invention also provides calibrant formulations that release mass calibrants in a slow, controlled manner.
