Abstract:
A flexible container arrangement useful for performing the magnetic separation of paramagnetic particles from a heterogeneous biological mixture. The flexible container arrangement of the invention includes two or more flexible containers (20, 22) having inlet and outlet ports. A flexible conduit (32) is connected between an outlet port of a first container and an inlet port of a second container. Another flexible conduit (32) is connected to an outlet port of the second container to allow for the discharge of fluid. The second flexible container (20, 22) is designed to provide for a reduction in fluid velocity at a rate of from about 1/7 to about 1/3 of the fluid velocity through the outlet conduit. The second container (20, 22) is also designed to provide a depth of fluid flow of about 0.02 to about 0.04 inch.

An apparatus and method for performing a magnetic cell separation process using paramagnetic beads, and having multiple steps or activitites which are required to complete the process successfully includes using a programmable machine (10) to either perform selected ones of the multiple steps itself; or to prompt a human attendant to perform others of said steps in sequence and to thereafter provide to the machine a verification input that the step to be performed by the human attendant is completed. The repeatability and reliability of the process when performed by the programmable machine (10) is vastly improved in comparison to conventional laboratory practice. Additionally, the programming of the machine (10) may be altered by super visory personnel to improve the efficacy of the process, or to customize the process for the separation of a particular type of cell.
Abstract:
A system for the selective separation of a specific cel population from a heterogeneous cell mixture is disclosed herein. Means are provided for obtaining a selective cell concentrate from the heterogeneous cell mixture by separating the selective cell concentrate based upon the physical or biological properties of the concentrate. This may be done by centrifugation, if desired. A container is provided having particle means, the particle means being attached to a substance that is active for binding to a specific desired cell. One brings the cell concentrate into contact with the particle means within the container, for example, by providing sterile connection between the site where the cell concentrate is formed and the container. This permits incubation of the cell concentrate with the particle means, to thereby selectively bind a specific cell population from the cell concentrate to the particle means, creating a particle/cell conjugate. Magnetic means are then provided for separating the particle/cell conjugate from the remainder of the cell concentrate. A two-magnet system may be used to form the separation.
