Abstract: PROBLEM TO BE SOLVED: To efficiently produce L-tertiary leucine useful for an asymmetric synthesis or the like of an amino acid derivative in good efficiency, high purity and high yield by carrying out an amino exchange of dimethyloxobutanoic acid (salt) in the presence of an amino acid by using a microorganism. SOLUTION: 3,3-Dimethyl-2-oxobutanoic acid (salt) is subjected to an amino exchange in the presence of an amino acid as an amino group donor by using a microorganism such as Alcaligenes faecalis DSM 4115, for example, at pH 5-9, preferably 7-8.5 to provide the objective L-tertiary leucine in the method for producing the objective L-tertiary leucine. Preferably, the amino group donor and 3,3-dimethyl-2-oxobutanoic acid are used in the ratio of (1/1)-(2/1), and the amino exchange reaction is carried out by using a cell-extract, an isolated total protein or a purified transaminase.

Abstract: Prodn. of L-tert.-leucine (Ia) or L-phosphinothricin (Ib) is effected by tranamination of 3,3-dimethyl-2-oxobutyric acid (IIa) or (3-carboxy-3-oxopropyl)methylphosphinic acid (IIb), or their salts, with an amino acid (III) as amino gp. binder. Pref. the reaction is effected in the presence of a transaminase enzyme at pH 7-8.5 using a (III):(II) ratio of 1-5:1, esp. 1-2:1. The enzyme is used in pure form, in the form of a microorganism, or in the form of a cell extract or protein isolate derived from a microorganism. The microorganism is a Paracoccus, Alcaligenes, Pseudomonas, Serratia, Agrobacterium, Streptomyces or Enterbacterium strain, or a genetically manipulated microorganism, esp. E.coli ATCC 11303 transformed with a plasmid contg. a tyrB or 1lvE gene.
