Described herein are compounds, methods, and kits for short- and long-term tracking of cell proliferation, differentiation, and/or function. The compounds disclosed herein are coumarin- based cell-tracking reagents that fluoresce in the blue portion of the UV/VIS spectrum and provide bright fluorescence intensity, uniform cell staining, good retention within cells and low toxicity toward cells.

Alkynyl-derivatized cap analogs, alkynyl-modified capped RNA, 1,4-disubstituted triazole-derivatized capped RNA, methods of preparation, methods of isolation, and uses thereof are provided. The "click" modification facilitates detection and isolation of capped RNAs and the 1,4-disubstituted triazole derivatives formed by the "click" reaction are useful for producing RNA transcripts and encoded protein.

The present disclosure provides methods, compositions and kits for measuring cellular nascent nucleic acid synthesis by colorimetric labeling of nucleic acid. The nucleic acid synthesis can be measured as cell proliferation, DNA or RNA synthesis, gene expression or apoptosis. Additionally, these methods may be used to screen compounds for their effect on cellular proliferation by treating cells or an organism with the test compound simultaneous to or before treatment with a competitive nucleoside analog.

Compositions and methods for detecting the activity of an ion channel in a cell are described. The methods include providing a loading buffer solution to the cell, where the loading buffer includes a thallium ion indicator and optionally chloride ions, and providing a stimulus buffer that includes thallium ions to the cell. Providing the stimulus buffer can cause thallium ion influx into or efflux out of the cell through the ion channel. After providing the stimulus buffer, a change in at least one optical property of the thallium ion indicator is detected in response to thallium influx or efflux, thereby detecting the activity of the ion channel.
