PURPOSE:An external preparation for the skin useful for remedying and preventing skin diseases such as humid tetter, acne, wound, etc., protecting the skin as a drug, quasi-drug, or cosmetic, containing 5-methylallantoin. CONSTITUTION:An external preparation for the skin containing 5-methylallantoin as an active ingredient. The external preparation for the skin has remedy promoting action on damaged parts such as wound, ulcer, burn, etc., astringent action, and covering action. It is useful for remedying erosion of the skin or mucosa, ulcer, and skin diseases such as humid tetter, etc., and used as a drug or quasi-drug having the above-mentioned medicinal effects. Since the active ingredient has no irritation to the skin, it may be blended with cosmetic, it prevents skin irritations of lips and chapped skin, is expected to have soft feeling in use, to make people undergo rejuvenation, etc. An amount of the active ingredient added is 0.01-20wt% based on the whole amount of pharmaceutical preparation, preferably 0.01-5wt% in the case of cosmetic, and 1-20wt% in the case of quasi-drug.

PURPOSE:To obtain the titled substance useful as pharmaceuticals and disinfectants, etc., safely in high yield and purity, by using formaldehyde or acetaldehyde as a raw material at a specific ratio to nitromethane, condensing the raw materials using an alkali, and brominating the product with bromine without separating from the reaction system. CONSTITUTION:1mol of nitromethane and >=1.5mol of formaldehyde or acetaldehyde are dissolved in a solvent and condensed with an alkali such as sodium hydroxide, potassium methoxide, etc. The condensation product is, without separating from the reaction system, brominated with bromine to obtain the compound of formula (R is H or methyl). The solvent used in the above reaction is water, lower alcohol, toluene, etc., and the condensation reaction is carried out at -20-+120 deg.C. Bromine may be used as it is in the bromination reaction, however, it is preferable to dissolve bromine in a solvent and add the solution dropwise to the reaction system. The amount of bromine is generally >=2 equivalent based on nitromethane.
