Abstract: An apparatus and method for detecting the type, presence or amount of bacteria in a sample. The apparatus and method involve staining a liquid sample with a fluorescent stain corresponding to bacteria and imaging one or more portions of the sample at a selected set of sample Z-axis heights, X-Y coordinates, or both, within the sample after a predetermined time to allow for non-bacterial components in the sample to settle out of solution. A calibration method for correlating the concentration or amount of bacteria in biological samples by different analysis methods including glass slide methods, agar plate colony culture methods and high precision methods.

Abstract: Rocky Mountain Spotted Fever (RMSF) is a vector borne disease that is caused by Rickettsia rickettsii infection. It is challenging to identify the disease in dogs and other mammals due to the cross-reacting antibodies from non-pathogenic Rickettsia spp. Provided herein are compositions and methods for detection and treatment of Rocky Mountain spotted fever (“RMSF”). The compositions specifically detect Rickettsia rickettsii and do not cross-react with other Rickettsia species providing rapid and accurate detection and diagnosis of RMSF.

Abstract: A method for calibrating sensitivity of a photometer includes measuring, by a double-beam spectrophotometer, an absorbance spectrum of a control solution, which has been diluted and includes a control substance. The method further includes linearly regressing the absorbance spectrum of the control solution over a predetermined range of wavelengths and determining whether a first slope of the linearly regressed absorbance spectrum of the control solution falls within a range of slopes of lines obtained from linearly regressing absorbance spectra of a plurality of reference solutions over the predetermined range of wavelengths. A concentration of chromophore in each reference solution is known and the absorbance spectra of the plurality of reference solutions have been obtained by the double-beam spectrophotometer.
