    Abstract: A fiber optic probe is provided with a distal sampling end, a proximal end, and light delivery and collection paths therethrough. The probe includes a window disposed at the distal sampling end of the fiber optic probe, the window having a distal end and a proximal end. A lens is disposed at the proximal end of the window, the lens having a distal end, a proximal end, and an aperture. A light delivery optical fiber is provided having a distal end and a proximal end, the distal end being received by the lens aperture. An optical isolator provided within the lens aperture to optically isolate the light delivery path and the light collection path. A collection optical fiber is provided in optical communication with the fiber collection filter. The probe may include a lens collection filter disposed between the window and the lens.

    Abstract: A method for obtaining one or more images of a sample using a microscope includes dividing, using a reverberation cavity, a first one of a plurality of laser pulses into a plurality of sequential sub-pulses, each of the plurality of sequential sub-pulses having a power that is less than a previous one of the plurality of sequential sub-pulses, directing, using the one or more lenses of the microscope, the plurality of sequential sub-pulses onto a portion of the sample to generate a plurality of signals, each of the plurality of signals being associated with a different depth within the sample, and detecting the plurality of signals from the sample to generate one or more images of at least a portion of the sample.

    Abstract: An apparatus for sorting cells includes a measurement volume that contains a cell to be measured, a light source that provides light to cause an emission by a fluorescent label attached to the cell, and an optic device that directs the light through the measurement volume. The apparatus flows the cells through the measurement volume such that as the cell flows through the measurement volume, it interacts with the light, resulting in a change in light originating from the measurement volume, the change in light is a fluorescence emission. Another optic device directs a portion of the light originating from the measurement volume to a detector, which detects the portion of the light. A processor operably coupled to the detector generates an estimate of DNA quantity in the cell based on the change in light originating from the measurement volume, and determines a characteristic of the cell from the estimate.

    Abstract: Systems and methods for characterizing biological specimens, which may involve identifying a cell type or state corresponding to a disease or health condition of a subject. A biological specimen is subjected to electromagnetic radiation for spectroscopic analysis such as Surface Enhanced Raman Spectroscopy (SERS) to determine the relative abundance of proteins or amino acids in the cells, which is used in a comparison to previously stored relative abundance data of a database to automatically identifies at least one of cell type and/or cell state of the cells (or the disease/health state of the subject with the disease state including the possibility of virus infection, or drug susceptibility of a subject to bacteria or fungus). The method may also be employed with biological entities or cellular structures such as exosomes and even protein or nucleic acid fragments to determine disease states or health states of the subject.
