Abstract: The present disclosure describes methods for analyzing mRNA poly(A) tail sequences with a high resolution to determine poly(A) tail length and heterogeneity. The method involves digesting an mRNA molecule to liberate a poly(A) tail, preparing a chromatographic sample comprising the mRNA poly(A) tails, preparing a second chromatographic sample comprising a reference sequence comprising a mRNA poly(A) tails having a predetermined length, separating the first and second samples by a chromatography method, which result in one or more chromatograms, and determining a sequence length of the mRNA poly(A) tails by comparing the chromatograms of the first and second samples. Chromatography methods for analyzing the poly(A) tail may include ultraviolet size-exclusion chromatography (SEC UV), ultraviolet ion-pair reversed-phase liquid chromatography (IP RP LC UV), ultra high performance liquid chromatography (UPHLC), and combinations thereof.

Abstract: A device for separating analytes is disclosed. The device has a sample injector, sample injection needle, sample reservoir container in communication with the sample injector, chromatography column downstream of the sample injector, and fluid conduits connecting the sample injector and the column. The interior surfaces of the fluid conduits, sample injector, sample reservoir container, and column form a flow path having wetted surfaces. A portion of the wetted surfaces of the flow path are coated with an alkylsilyl coating that is inert to at least one of the analytes. The alkylsilyl coating has the Formula I:

Abstract: Methods are provided for making rapid labeled dextran ladders and other calibrants useful in liquid chromatography. The methodologies include a two-step process comprising a reductive amination step of providing a reducing glycan and reacting it with a compound having a primary amine to produce an intermediate compound. The intermediate compound is then rapidly tagged with a rapid tagging reagent to produce the rapid labeled dextran ladder.

Abstract: Methods for quenching a nuclease digestion of a target nucleic acid prior to downstream analysis of the target nucleic acid are disclosed herein. Particularly, methods for controlling the end point of a nuclease digestion prior to sequence analysis of a target nucleic acid is provided. Quenching of a nuclease digestion in the present disclosure employs at least one non-ionic or anionic denaturant combined with an optional reducing agent. The methods presented in this disclosure aids preserving the sample comprising the target nucleic acid or fragments thereof for long term storage and ensures that the effect of contaminating nucleases is eliminated during pretreatment step.
