Abstract: Disclosed herein are methods and compositions for the cryopreservation of stem cells, such as stem-cell derived retinal pigment epithelial cells, that have been seeded onto and cultured on a substrate, such as a polymeric substrate. Such cryopreserved stem cells are useful for cell therapies, such as treatment of ocular damage or disease.

Abstract: Methods and compositions modulate the activity of electrically excitable cells. Photovoltaic compounds which, upon exposure to light energy, increase or decrease the electrical activity of cells. These supplement and/or replace of vision based on the conversion of light energy to electrical energy within certain cells of the visual system. A “patch” or bridge to circumvent one or more defective, damaged, or diseased cells in the visual system. Additionally, in several embodiments, subjects with normal vision can benefit from the methods, compositions, systems, and/or devices disclosed herein as normal visual acuity can be heightened. The exposure induces an energy (e.g., a receipt of light energy, conversion to electrical energy, and passage of that electrical energy) from the photovoltaic compound to the cell, thereby altering the transmembrane potential of the cell and/or the opening of one or more ion channels, thereby modulating the activity of the electrically excitable cell.

Abstract: A medical device having a permeable bag connected by a non-permeable cannula to a discharge sac is described along with a manufacturing process and surgical implantation method. The permeable portions of the device have pores that are sized to be permeable to a predetermined class of small molecules, such as oxygen, nitrous oxide, or other therapeutic agents. Once absorbed inside the device, the small molecules are then passively transported, by a concentration gradient of the small molecules, to the discharge sac to be disbursed. A metal tube or other strip can be included in the cannula to assist a surgeon in orienting the device within the body.

Abstract: A contact lens fluid delivery device having a liquid reservoir connected to a channel with a flow regulator is described. Other eye hydration and variable dioptric power contact lenses are described herein. Also described are implantable liquid delivery apparatuses having a liquid storage reservoir connected to a channel with a flow regulator. These devices and apparatuses are useful for specific, targeted delivery of therapeutic liquids within a subject. In some embodiments, the devices incorporate actuation chambers which provide a driving force releasing the fluid into the targeted area e.g., the eye. The actuation chambers described herein can contain phase change materials or osmotic chambers or a combination thereof to drive the release of fluid.

Abstract: A microfluidic flow restrictor that uses micron-sized beads to impede flow is described. The flow rate can be adjusted by adding or removing the beads using injection needles through self-sealing ports, one injection needle injecting or aspirating beads and another injection needle pushing or pulling fluid from outside of a bead trap within the flow restrictor. In alternative embodiments, the beads or other filler material can be trapped in a manifold bead trap such that they block a subset of fluid channels of the flow restrictor, allowing fluid to flow freely through the rest of the fluid channels. The flow restrictor can be integrated with a contact lens or implantable medical device for use in dispensing liquid therapeutic agents at flow rates of microliters per minute or moving body fluids at a controlled rate from one part of the body to another.

Abstract: A system and method are disclosed for causing or enhancing the perception of color in a blind or visually impaired person. The system may comprise: an electromagnetic stimulation device associated with the blind or visually impaired person; and a visualization device for receiving an image and detecting a color of an object in the image, wherein the electromagnetic stimulation device is configured to stimulate the blind or visually impaired person based on a parameter of electromagnetic stimulation that varies based on the color detected by the visualization device. The method may comprise: varying a parameter of electromagnetic stimulation of an electromagnetic stimulation device, associated with an eye of the blind or visually impaired person, based on which color is detected by a visualization device that is used in conjunction with the electromagnetic stimulation device; and stimulating the eye with the electromagnetic stimulation device according to the parameter of electromagnetic stimulation.
