Abstract: The present teachings relate to a method of making a biological sample collection container, an internally coated biological sample collection container, and uses of the same, particularly for omic analysis. A reagent (or a reagent precursor) is deposited in a liquid state at least partially along at least one side wall of the container. The reagent precursor is dried to form a dried coating having a predefined pattern and topology along at least a portion of the at least one side wall. A container thus results having a coating that includes, in a dried state, a stabilizer agent, or reaction product of a stabilizer agent and an anticoagulant, and upon collection of a sample enables stabilization of any present white blood cells, cell-free nucleic acids, extracellular vesicles, circulating tumor cells, proteins, metabolites, lipids, or any combination thereof, and preserving them in sufficient quantity and quality for omic analysis.

Abstract: Devices including a composition for preserving and processing cell-free nucleic acids located within a blood sample and methods of using the same are disclosed, wherein a blood sample containing cell-free nucleic acids is treated to reduce both blood cell lysis and nuclease activity within the blood sample. The device may include a direct blood draw tube comprising a composition formulated for stabilizing cell-free nucleic acids within a blood sample including one or more formaldehyde releaser preservative agents, ethylenediaminetetraacetic acid (EDTA), one or more solvents, and formaldehyde. The composition is free of separately added formaldehyde but contains the formaldehyde as a result of the one or more formaldehyde releaser preservative agents. The treatment of the sample aids in increasing the amount of cell-free nucleic acids that can be identified and tested while maintaining the structure and integrity of the nucleic acids.

Abstract: Provided herein are methods of preparing a protein sample for proteomic analysis. In exemplary embodiments, the method comprises (a) contacting a blood sample comprising proteins with a protective agent comprising an anticoagulant (AC) and an aldehyde releaser (AR), to obtain a mixture, optionally, wherein the blood sample is added to a blood collection tube (BCT) comprising the protective agent, and (b) isolating a fraction comprising proteins or a source of proteins from the mixture to yield a protein sample or a source of a protein sample, wherein steps of the method are carried out in the absence of exogenous proteolytic enzyme inhibitors, wherein the protein sample is suitable for proteomic analysis. Preferably, the protective agent consists of essentially (i) about 300 g/l to about 700 g/l imidazolidinyl urea; (ii) about 20 g/l to about 60 g/l glycine; and (iii) about 60 g/l to about 100 g/l EDTA; and the protein sample is analysed via mass spectrometry-based proteomic methods.

Abstract: The claimed subject matter comprises a device to collect and preserve cells comprising of: (1) a collection container comprised of a tube having an open end and a closed end, a closure in the open end of the tube, a vacuum drawn to a predetermined level inside the container; and (2) compounds including an anticoagulant agent and a fixative agent, wherein the compounds are in a sufficient amount to preserve said cells” original morphology and antigenic sites without significant dilution of said cells, and thereby allowing said cells to be directly analyzed by a flow cytometer without further treatment. The claimed subject matter further comprises of a method of making a collection device for cells comprising of: (1) providing a tube having an open end and a closed end; (2) preloading compounds including: an anticoagulant agent, and a fixative agent into the tube, wherein the compounds are in a sufficient amount to preserve the cells” original morphology and antigenic sites without significant dilution of the cells, and thereby allowing the cells to be directly analyzed by a flow cytometer without further treatment; (3) inserting a closure into the open end of the tube; and (4) drawing a vacuum inside the tube to a predetermined level to form the collection device.
