Abstract:
PURPOSE: Provided is an isolation method of a secondary metabolic product with herbicidal effect on the white clover(Trifolium repens), which is released from culture extract of isolate BWC98-105(Sclerotinia sp.), thereby withering the subterranean stem and stem of the white clover continuously, and inhibiting the generation thereof consequently to reduce the consumption of agricultural chemicals. CONSTITUTION: An isolation method of a secondary metabolic product with herbicidal effect on the white clover(Trifolium repens) by using BWC98-105(Sclerotinia sp.) comprises the steps of: culturing BWC98-105 strain in PD broth medium for 7 days, followed by systematic extraction with an organic solvent; fractioning the extraction with butanol and ethylacetate; separating the butanol and ethylacetate fractions using the column filled with reverse phase silica gel(C18); and analyzing the separated fractions by HPLC, wherein a second metabolic product having herbicidal effect on the white clover(Trifolium repens) is produced at retention time intervals of 4.13 minute.

PURPOSE: A non-destructive analysis method is provided to effectively analyze ingredient consisting of fatty acid of a grain. CONSTITUTION: A non-destructive analysis method for a fatty acid composition of a single granule comprises: a step of randomly selecting a plurality of prediction model development group and prediction model evaluation group; a step of measuring near infrared absorption spectrum of two groups; a step of randomly selecting grain granule sample among the prediction model development groups and measuring near infrared ray absorption spectrum; a step of pulverizing the granule sample and performing chemical analysis of fatty acid ingredients; and a step of applying primary and secondary derivatives to the absorption spectrum.

PURPOSE: A method for non-destructive analysis of one seed grain by near infrared ray absorption spectrum is provided to precisely analyze the components and characteristics of one seed grain in a short time without using any additional chemical analysis equipments. CONSTITUTION: A method for non-destructive analysis of one seed grain by near infrared ray absorption spectrum includes the steps of measuring near infrared ray absorption spectrums(400-2500mm) of 30-100 seeds of selected grain samples by a horizontal manual module, correcting absorbancy per wavelength measured by 30-100 grains respectively with a single average absorption spectrum per wavelength, randomly distinguishing 150 averaged absorption spectrum into about 100 estimation model developing models and about 50 estimation model testing models, correcting the uniformity of the samples by using a sieve of 60 mesh after preparing powdered samples, testing the moisture content of the powdered samples by drying for 2 hours in a drier of 105°C, precisely carrying out chemical analysis of contents of components such as protein and fat contained in the powdered samples twice or more, removing samples of which a difference in the chemical analysis is beyond a range of 0.2-0.4% from the models, inserting real component contents of which moisture contents are corrected into the measured average absorption spectrum, carrying out the statistic analysis for setting optimum estimation models for the respective components, and selecting the optimum estimation models by discriminating the precision of the estimation models.

본 발명은 클로버에 대하여 기주특이적으로 기생하고 제초활성을 갖는 스클레로티니아 트리폴리오럼 BWC98-105 균주를 이용하여 이 균주가 분비하는 클로버 제초활성 2차 대사산물(secondary product)을 분리하는 방법에 관한 것으로, 스클레로티니아 트리폴리오럼 BWC98-105 균주(KACC93004)를 PD Broth 배지에서 7일 동안 액체 배양하는 단계; 상기 배양액을 여과하는 단계; 상기 여과된 배양액을 헥산, 클로로포름, 에틸아세테이트 및 부탄올 순으로 액체분배추출하고, 에틸아세테이트 및 부탄올 각각의 분획물을 혼합하는 단계; 상기 분획 혼합물을 역상 실리카겔(C 18 )로 충진된 컬럼으로 분리하는 단계; 및 상기에서 분리된 분리물을 고속액체색층분석기(HPLC)를 이용하여 4.13분의 머무름 시간에 추출되는 화합물을 수득하는 단계;를 포함하여 이루어진다. 본 발명의 분리방법으로 얻어지는 대사물질은 신속히 클로버 지하경 및 줄기 등 지상부 식물체를 고사시키고, 다음해의 잔디 포장을 오염시키는 클로버의 발생을 억제시키는 효과가 있다.
