    Abstract: PURPOSE: An expression vector for human CD 73, a cell line and a transgenic-cloned animal including the same are provided to be able to produce mini pigs for organ transplantation with a high value added by developing a novel immune rejection response adjusting technology, and to be able to accelerate practical use of xenotransplantation by reproducing transgenic-cloned pigs for organ transplantation. CONSTITUTION: An expression vector includes polynucleotide coding the CD73 gene, a mini pig endothelial cell specific promoter. A manufacturing method of a transgenic pig comprises 1) a step of isolating a somatic cell from a pig; 2) a step of manufacturing an expression vector and introducing the expression vector to the somatic cell; 3) a step of selecting and cultivating the clone somatic cell with the expression vector introduced in step 2); 4) a step removing the nucleus of the ovum collected from sow, and fusing with the clone somatic cell of step 3); and 5) a step of transplanting the fused cloned egg to surrogate sow and reproducing weaners.     Abstract: PURPOSE: A gene targeting knock-in vector including protein over-revelation cassette, a manufacturing method thereof, and a transformation cloned animal for heterograft in which the vector is introduced are provided to manufacture a transformation reproduction miniature reproduction pig in which a graft rejection is controlled by effectively knock-out and knock-in at the same time. CONSTITUTION: A gene targeting knock-in vector successively including protein over-revelation cassette comprises a first area having base sequences which are homologous in gene of mammal, a cassette for protein over-revelation, and a second area having homologous base sequences in gene of mamma. The gene of mammal is alpha 1,3- galactosyltransferase gene. The mammal is a pig. The pig is a NH-miniature pig. The first area is a left arm including a part of intron 8 of the alpha 1,3- galactosyltransferase or Exxon 9. A size of the first area is 2kb~6kb.
