Abstract:
PURPOSE: A cellobiohydrolase isolated from Fomitopsis pinicola is provided to ensure excellent thermal stability and activity at high temperature. CONSTITUTION: A method for preparing Fomitopsis pinicola-derived cellobiohydrolase comprises: a step of centrifuging culture liquid of Fomitopsis pinicola and washing; a step of mixing a washing solution and supernatant and filtering to prepare coenzyme liquid; and a step of purifying the coenzyme liquid by column chromatography. The cellobiohydrolase contains a Lys-C(LYS) fragment and trypsin fragment.
Abstract:
An endoglucanase derived from fomitopsis palustris is provided to develop wood-biomass resolving system and manufacturing technique of wood-biomass lysis enzyme. An endoglucanase EG47 derivated from fomitopsis palustris contains amino sequence ALFGLMNEPH and AAGAWNYLLL, molecular weight is 47kDa. The endoglucanase has microcrystal cellulose lysis activity. Wood-biomass lysis enzyme comprises Endo-glucanase EG 47 or Endo-glucanase EG 35. The studying for the Endoglucanase comprises: a step of purifying glycosylhydrolase from fomitopsis palustris; a step of analyzing enzyme and concentrating protein; a step of identifying purified enzyme by analyzing amino sequence; a step of analyzing specificity of purified Endoglucanase; and a step of analyzing with chromatography.

본 발명은 바이오필름(biofilm) 형성을 조절하는 물질을 스크리닝하는 방법에 관한 것으로서, 보다 상세하게는 벼의 흰잎마름병의 원인균인 잔토모나스 오리제( Xanthomonas
oryzae )의 바이오필름 형성 조건을 사용하여 미생물의 바이오필름 형성 조절 물질을 대량으로 신속하고 정확하게 스크리닝하는 방법에 관한 것이다. 본 발명의 바이오필름 형성 조절 물질 스크리닝 방법은 기존의 방법에 비하여 정확성, 간편성, 신속성에서 우수한 특징이 있으므로 다량의 바이오필름 형성 조절 후보물질을 단시간 내에 정확하게 스크리닝할 수 있다.
