Abstract:
PURPOSE: An extracellular enzyme which is prepared by culturing Fomitopis palustris in a culture medium containing ligneous biomass, especially rice straw, a method for producing the same, and a method for saccharifying the biomass are provided to enhance cellulase activity. CONSTITUTION: A saccharifying method for producing bioethanol comprises: a step of adding Fomitopis palustris-derived extracellular cellulase into a culture medium containing a rice straw substrate as a carbon source; and a step of saccharifying at 45 deg. C and pH 4.5-5.0 or adding Tween20. A saccharified material for producing bioethanol is prepared by the method. The saccharifying ability of the Fomitopis palustris-derived extracellular cellulase is improved in rice straw with low crystallinity of cellulose.
Abstract:
PURPOSE: A Porodisculus pendulus-derived diastatic enzyme and a method for producing biofuel using the same are provided to ensure excellent saccharification performance. CONSTITUTION: A method for producing Porodisculus pendulus-derived diastatic enzyme comprises: a step of culturing Porodisculus pendulus strain in a culture medium containing cellulose, corn powder, or peptone/yeast extract mixture at 25-30°C and pH 5.5-6.0; and a step of isolating the diastatic enzyme from the culture liquid. A used biomass is cellulose or rice straw.
Abstract:
PURPOSE: A cellobiohydrolase isolated from Fomitopsis pinicola is provided to ensure excellent thermal stability and activity at high temperature. CONSTITUTION: A method for preparing Fomitopsis pinicola-derived cellobiohydrolase comprises: a step of centrifuging culture liquid of Fomitopsis pinicola and washing; a step of mixing a washing solution and supernatant and filtering to prepare coenzyme liquid; and a step of purifying the coenzyme liquid by column chromatography. The cellobiohydrolase contains a Lys-C(LYS) fragment and trypsin fragment.
