A method for preparing Scleroglucan by cultivating Sclerotium sp. in a culture medium including mandarin peels as a carbon source is provided to use the mandarin peels economically, thereby reducing manufacturing costs for the culture medium. A method for preparing Scleroglucan by cultivating Sclerotium sp. in a culture medium including mandarin peels as a carbon source comprises the following steps: saccharifying the mandarin peels with acids; adding a diastatic enzyme to the saccharified mandarin peels so as to saccharify the mandarin peels additionally; cultivating Sclerotium rolfsii in the culture medium wherein the saccharified mandarin peels are obtained; and collecting the Scleroglucan from the cultivated Sclerotium rolfsii.

PURPOSE: A liquid culture medium composition for culturing inonotus obliquus mycelia is provided to mass-produce protein-bound polysaccharide having an immunity enhancement effect. CONSTITUTION: A liquid culture medium composition for culturing inonotus obliquus mycelia contains 8~16g/L of glucose, 20~32g/L of fructose, and 4~8g/L of soytone peptone. The culturing method of the inonotus obliquus mycelia on the liquid culture medium composition comprises the following steps: passage culturing the inonotus obliquus mycelia on an agar medium; static culturing the inonotus obliquus mycelia on a stationary culture medium; and liquid culturing the inonotus obliquus mycelia on a liquid medium.

CONSTITUTION: A culture medium composition for cultivating inonotus obliquus mycelia contains 22g of glucose, 30g of oil, 6g of casein, 0g of soytone peptone, 3g of KH2PO4(calcium hydrogen phosphate), and 1L of distilled water. The oil is oleic acid. The inonotus obliquus mycelia are passage cultured on an agar medium, static cultured on a static cultivation medium, and liquid cultured on a liquid medium.

A method for culturing a Sparassis crispa mycelia is provided to massively produce Sparassis crispa mycelia and shorten culture time through semi-continuous culture of liquid phase. A medium for culturing Sparassis crispa mycelia in liquid phase comprises 0.01~3.0% of glucose, 0.01~0.5% of yeast extract, 0.01~0.2% malt extract, 0.01~1% of potassium monophosphate, and 0.01~0.04% of iron sulfate. A method for culturing the 0.01~0.04% in liquid phase comprises: a step of germinating a spore of Sparassis crispa mycelia; a step of culturing in liquid phase medium at 23‹C, 200 rpm of shaking speed for seven days; and a step of collecting beta-glucan from Sparassis crispa mycelia.
