Abstract:
본발명은재조합벡터로형질전환된스테비올바이오사이드생산용재조합미생물및 상기재조합미생물을이용한스테비올바이오사이드의생산방법에관한것이다. 본발명에따른재조합벡터로형질전환된재조합미생물은스테비아의감미성분으로서스테비올의배당체인스테비올바이오사이드를다량으로생산하는우수한효과가있다. 따라서, 본발명의재조합미생물을이용할경우, 스테비아의추출물로부터직접스테비올바이오사이드를분리하는것이아닌, 미생물에도입된재조합유전자에의해대사경로를조작함으로써, 간단한공정만으로도스테비올바이오사이드를대량으로제공할수 있어, 이를이용하는천연감미료, 음료등의식품산업에유용하게이용될수 있다.
Abstract:
PURPOSE: Recombinant E. coli with reconstructed sucrose metabolic pathways is provided to be used in a microorganism-based industry by transforming E.coli which produces a secondary metabolite using only sucrose as a carbides source. CONSTITUTION: Sucrose-using E.coli is a reconstructed PTS(phosphoenolpyruvate (PEP)-dependent phosphotransferase transport system) system or non-PTS system. The PTS system is prepared by transduction of sucrose phosphotransferase, sucrose hydrolase, and fructokinase. The non-PTS system is prepared by transduction of sucrose permease, sucrose hydrolase or phophorylase, and fructokinase. The PTS-syste-introduced E.coli is prepared by transformation with pAC-AAK plasmid which expresses scrA, sacA, and sack genes. The gene is derived from Staphylococcus xylosus and Lactobacillus plantarum.

PURPOSE: A recombinant microbe with menaquinone productivity, and a method for preparing menaquinone using the same are provided to induce excessive production of menaquinone. CONSTITUTION: A recombinant microbe with menaquinone productivity over-expresses one gene selected from the group consisting of UbiE(ubiquinone-8/menaquinone-8 biosynthesis methyltransferase), MenA(1,4-dihydroxy-2-naphthoate octaprenyltransferase), MenB (naphthoate synthase), MenC(osuccinylbenzoate-CoA synthase), MenD(2-succinyl-6-hydroxy-2,4-cyclohexadiene-1-carboxylatesynthase/2-oxoglutarate decarboxylase), MenE(o-succinylbenzoic acid-CoA ligase), MenF(menaquinonespecificisochorismate synthase), ispA (geranyltranstransferase), dxr(1-deoxy-D-xylulose 5-phosphatesynthase), and idi(isopentyl diphosphate isomerase). A method for preparing menaquinone comprises a step of culturing the recombinant microbe, and a step of collecting menaquinone from a culture medium of the recombinant microbe.

PURPOSE: A shuttle vector comprising plasmid from lactic acid bacteria is provided to stably express a gene or protein and to be able to reproduce over 100 generations due to a stable method of theta-typed reproduction. CONSTITUTION: Plasmid pMBLT00 comprises the sequence number marked sequence number 1(SEQ ID NO:1). Plasmid pMBLT00 is separated Leuconostoc mesenteroides. A shuffle vector capable of reproduction both in LAB-E-coli and lactic acid bacteria comprises sequence number marked the sequence number 1 and produced by using plasmid pMBLT00 and transformation vector from LAB-E-coli. The transformation vector from LAB-E-coli is pUC19. The shuffle vector has a method of theta-typed reproduction. A transformant transformed from the shuffle vector is LAB-E-coli or lactic acid bacteria. Lactic acid bacteria are Lactococcus, Leuconostoc or Pediococcus. A method of reproduction comprises the following steps. The shuffle vector capable of reproduction both in LAB-E-coli and lactic acid bacteria is made by using plasmid pMBLT00 with the sequence number marked the sequence number 1 and transformation vector from LAB-E-coli(a). The recombinant vector is made combining the shuffle vector comprising a target gene which codes target proteins(b). The recombinant vector is injected into microorganism.
