Abstract:
A method for screening and detecting a swine fever virus antibody is provided to easily screen and diagnose infected swine fever virus antibody by using a monoclonal antibody specifically reacting with a recombinant swine fever virus Erns protein and secure constant sensitivity regardless of all genotypes of the swine fever virus when using all three kinds of recombinant protein. A recombinant swine fever virus Erns is prepared by expressing a gene encoding three Erns proteins among swine fever viruses having various genotypes using baculovirus. A method for preparing a monoclonal antibody specifically reacting to the Erns comprises the steps of: (a) constructing a swine fever virus LOM strain-derived Erns recombinant gene using a baculovirus expression system; and (b) producing the monoclonal antibody using the recombinant Erns protein. A method for screening and detecting a swine fever virus antibody comprises the steps of: (a) mixing three kinds of Erns recombinant proteins at proper concentration to prepare an antigen cocktail; (b) after diluting the antigen cocktail into a coating buffer solution, pouring it in a plate to be absorbed into the plate; (c) washing the recombinant antigen not-absorbed into the plate to remove it; (d) adding a sample to be tested to the plat to allow the reaction; (e) washing the sample to be tested not specifically bound to the recombinant Erns antigen protein absorbed into the plate to remove it; (f) adding a monoclonal antibody not being bound to the enzyme but being specifically reacted to the Erns protein to allow the reaction; (g) washing the monoclonal antibody not being bound to the recombinant Erns antigen protein absorbed into the plate to remove it; and (h) adding a substrate reacting with the enzyme to determine the swine fever virus infection.
Abstract:
PURPOSE: Provided are a genetic recombinant baculovirus and a recombinant spike protein of transmissible gastroenteritis virus(TGEV) manufactured therefrom. And a monoclonal antibody capture enzyme linked immunosorbent assay for detecting the antibody of TGEV using the same protein is also provided, thereby the antibody of TGEV can be rapidly detected. CONSTITUTION: The recombinant spike protein of TGEV is produced by the steps of: isolating and identifying TGEV from pigs died of diarrhea; analyzing the nucleotide sequence of spike protein of TGEV; inserting the TGEV spike gene into the baculovirus gene to produce a recombinant baculovirus(KFCC-11015); infecting the recombinant baculovirus into SF9 cells to express TGEV spike protein. The antibody of TGEV is produced by inoculating the expressed recombinant TGEV spike protein into Guinea pigs. The monoclonal antibody capture enzyme linked immunosorbent assay using TGEV spike protein that is produced from the recombinant baculovirus(KFCC-11015) in Guinea pigs as an antigen is used in detecting the neutralizing antibody of TGEV.

본 발명은 -20℃ 이하에서 동결되지 않으면서, 구제역바이러스에 대한 소독효과를 갖는 구제역바이러스 소독용 조성물에 관한 것으로, 구연산 또는 구연산을 유효성분으로 함유하는 소독제, 및 동결방지제를 포함한다. 본 발명의 구제역바이러스 소독용 조성물에서, 상기 동결방지제는 메탄올을 유효성분으로 함유하는 자동차유리 세척제, 에틸렌글리콜, 프로필렌글리콜, 염화나트륨, 염화칼슘 및 에탄올로 이루어진 군으로부터 선택되는 1종 이상일 수 있다.

본 발명은 구제역 A형 한국 분리 바이러스(A/KOR/Pocheon/2010)의 전체 유전자 부위 중에서, 상기 A형 한국 분리 바이러스의 방어 항원인 P1 단백질을 코딩하는 유전자가, 구제역 A형 중동 유래 아시아 지역형 바이러스(A/Iran/05)의 방어 항원인 P1 단백질을 코딩하는 유전자로 치환된 재조합 유전자가 삽입된 플라스미드를 포함하는 재조합 구제역 바이러스 및 그의 제조방법에 관한 것이다.
Abstract:
A mixed live vaccine of transmissible gastroenteritis virus, porcine epidemic diarrhea virus and porcine rotavirus, and a vaccination method by using the same live vaccine are provided to prevent diarrhea of pigs at one time, reduce vaccination time, and minimize titer loss of attenuated viruses by using trehalose. The mixed live vaccine is prepared by mixing a transmissible gastroenteritis virus(KCTC 10889BP), a porcine epidemic diarrhea virus(KCTC 10891BP) and a porcine rotavirus(KCTC 10890BP) in a weight ratio of 1:1:1, and adding a trehalose phosphate glutamate gelatin mixture. The mixed live vaccine is vaccinated by vaccinating a mother pig with the mixed live vaccine one time, and vaccinating the first vaccinated mother pig with the vaccine of transmissible gastroenteritis virus(KCTC 10889BP) and porcine epidemic diarrhea virus(KCTC 10891BP) two times.
