Abstract: PURPOSE: A vitrification solution and loading solution are provided to enhance high usage and to improve viability and regeneration rate. CONSTITUTION: A vitrification solution contains 42.5 of glycerol, 15 of dimethylsulfoxide, 15 of ethylene glycol, and 17.5 of sucrose; 37.5 of glycerol, 15 of dimethylsulfoxide, 15 of ethylene glycol, and 22.5 of sucrose; 32.5 of glycerol, 15 of dimethylsulfoxide, 15 of ethylene glycol, and 27.5 of sucrose; and 35.0 of glycerol, 15 of dimethylsolfoxide, 15 of ethylene glycol, and 20.0 of sucrose (%, w/v). A loading solution contains 9.2 of 1.0 M glycerol, 23.9 of 0.70M of sucrose, 18.4 of 2.0 M glycerol and 22.25 of 0.65M sucrose; and 15.0 of 1.63M glycerol, 15.0 of 0.44M sucrose, 17.5 of 1.9M glycerol, and 17.5 of 0.51M of sucrose.

Abstract: PURPOSE: A cryopreservation and regeneration method for Chrysanthemum shoot is provided to preserve Chrysanthemum using vitrification solution and loading solution. CONSTITUTION: A cryopreservation for cryopreservation shoot comprises: a step of extracting laterial bud of in vitro cultured cryopreservation; a step of pre-culturing in 0.3M-0.5M-0.7M of sucrose medium; a step of culturing in a loading solution(17.5% of glycerol and 17.5% of sucrose) for 40-50 minutes; a step of shaking and dehydrating the Chrysanthemum; and a step of rapidly-freezing in the liquid nitrogen.
