Abstract: PURPOSE: A transgenic mouse which expresses luciferase and a method for producing the same are provided to effectively detect allergen existing in an environment. CONSTITUTION: A method for screening allergen comprises: a step of treating allergen to a transgenic mouse which is produced from transgenic mouse fertilized egg(deposit number KCTC 11575BP) for detecting allergen; and a step of measuring the amount of luciferase expressed in the transgenic mouse. A method for manufacturing mouse pIL-4/Luc/CNS-1 expression cassette expressing luciferase comprises: a step of amplifying IL-4 promoter base sequence of sequence number 1 by PCR; and a step of constructing pIL-4/Luc/CNS-1 vector by binding pTransLucent vector and amplified IL-4 promoter by restriction enzymes, NheI and BglII.

Abstract translation: 目的：提供表达荧光素酶的转基因小鼠及其生产方法，以有效地检测环境中存在的过敏原。 构成：筛选过敏原的方法包括：对转基因小鼠进行变应原的步骤，该转基因小鼠由转基因小鼠受精卵（保藏号KCTC 11575BP）产生，用于检测变应原; 以及测量转基因小鼠中表达的荧光素酶的量的步骤。 用于制造表达荧光素酶的小鼠pIL-4 / Luc / CNS-1表达盒的方法包括：通过PCR扩增序列号1的IL-4启动子碱基序列的步骤; 以及通过用限制酶NheI和BglII结合pTransLucent载体和扩增的IL-4启动子来构建pIL-4 / Luc / CNS-1载体的步骤。

Abstract: PURPOSE: A transgenic animal which expresses human GATA3 gene under the regulation of a cytomegalovirus promoter is provided to study the function of GATA3 protein in vivo and correlation between GATA3 protein and allergic disease. CONSTITUTION: A transgenic fertilized egg(deposit number KCTC 12001BP) is prepared by transforming human GATA3 gene of sequence number 1. A transgenic mouse is produced by the fertilized egg. An expression cassette has human GATA3 gene which is linked with a cytomegalovirus promoter. A method for preparing a pCMV/hGATA3 expression cassette comprises: a step of amplifying a human GATA3 base sequence by PCR; and a step of cloning to pcDNA3.1 V5-His-TOPO vector.

Abstract translation: 目的：提供在巨细胞病毒启动子调控下表达人GATA3基因的转基因动物，以研究GATA3蛋白在体内的功能和GATA3蛋白与过敏性疾病之间的相关性。 构成：通过转化序列1的人GATA3基因制备转基因受精卵（保藏号KCTC 12001BP）。受精卵产生转基因小鼠。 表达盒具有与巨细胞病毒启动子连接的人GATA3基因。 制备pCMV / hGATA3表达盒的方法包括：通过PCR扩增人GATA3碱基序列的步骤; 和克隆到pcDNA3.1 V5-His-TOPO载体的步骤。
