PURPOSE: A polypeptide containing Oct-4 binding domain of PKM2(pyruvate kinase type M2) is provided to control cell differentiation by promoting or suppressing Oct-4 transcription. CONSTITUTION: A polypeptide promotes Oct-4 transcription factor activity by binding to the Oct-4. The polypeptide is an Oct-4 binding domain of PKM2 protein comprising amino acid of the sequence number 2. An expression vector for regulating Oct-4 transcription factor activity contains a polynucleotide encoding the polypeptide or anti-sense polynucleotide which is complementary to the polynucleotide.

A method for screening antineoplastic comprises the following steps of: treating candidate antineoplastic in a host cell transformed by a vector containing EWS-Oct-4 fusion protein coding polynucleotide sequence operately linked to a promoter and a vector containing a reporter gene operately linked to a promoter positively controlled by the EWS-Oct-4 fusion protein; and measuring degree of expression of the reporter gene in the transformed host cell. The EWS-Oct-4 fusion protein has amino acid sequence of SEQ ID NO:1.
