Abstract: PURPOSE: Thermococcus pacificus DNA polymerase variant is provided to be used in a various way such as gene diagnosis and PCR reaction thanks to being high in DNA amplification and amplification speed during PCR. CONSTITUTION: DNA polymerase comprises amino acid sequences chosen from a group comprised of sequence number 9, 10 and 11. A recombinant vector comprises amino acid sequence coding the amino acid sequences above. A host cell is transformed into the recombinant vector. The method of manufacturing DNA polymerase comprises the following steps. A recombinant vector which comprises base sequence s coding the amino acid is manufactured. The recombinant vector is transformed into a host cell. The transformed cell is cultivated. Protein from the transformed cell is collected.
