Abstract: Pyrimidine derivs. of formula (I) are new. In (I) R1, R2 and R3 are each H, Cl, CH3 or CH3O, R4 and R5 are each CN, 2-7C alkoxycarbonyl, (I) carbanilide or carboguanidide, 2-5C aliphatic acyl, benzoyl (opt. ring substd. by Cl, CH3 or CH3O), 1-6C alkyl, allyl, phenyl (opt. substd. as benzoyl) or furyl, R4+R5 may together complete an aliphatic mono- or poly-carbocyclic or a heterocyclic ring each with 5-6 ring members, opt. substd., and with >=1 carbonyl gp. alpha to the C atom shown. Alternatively, one of R4 and R5 = H and the other is opt. substd. benzoyl. They are made by reacting the corresp. 2,4-diaminopyrimidine with R6O.CH=CR4R5 (R6 is H or 1-3C alkyl). (I) are antibacterials and esp. potentiators for sulphonamides. They are formulated conventionally at (I): sulphonamide ratio 1:10-5:1(1:1-5).

Abstract: Pyrimidine deriv. of formula (I) is new. (I) is prepd. by reacting trimethoprim with a dichloroacetic acid deriv. at 20-150 degrees C, opt. in presence of solvent and acid acceptor. (I) is an antimicrobial and also potentiates the activity of sulphonamides. It is formulated conventionally for treating e.g. throat, nose, ear, respiratory, digestive or urinary tract infections. It is more active in vivo that trimethoprim e.g. 2.7 times more active against Streptococcus agalactiae in mice.
