Abstract:
It is intended to clarify the undisclosed part of the mechanism controlling the circadian rhythm. It is newly found out that RORa (retinoic acid binding-receptor alpha, the same applies hereinafter) promotes the induction of Bmal1 expression and that the induction of Bmal1 expression is promoted under hypoxic conditions. These facts strongly suggest the presence of the mechanism controlling the circadian rhythm wherein, when RORa expression is promoted under hypoxic conditions etc., the induction of Bmal1 expression is promoted and the promotion of the induction of Bmal1 expression promotes the binding of BMAL1 to CLOCK to thereby promote the induction of the expression of the Per gene or the Cry gene. Accordingly, the above findings are likely applicable to agents for regulating jet lag and anticancer drugs.

PROBLEM TO BE SOLVED: To provide a method for determining a gene expression amount using a proper control gene in a method for determining a gene expression in a hair follicle cell. SOLUTION: The method for determining a gene expression is a method for determining the expression amount of an objective gene in a hair follicle cell, which comprises using one or more genes selected from a fixed gene described in a sequence listing and its homolog as a control gene. PROBLEM TO BE SOLVED: To provide a method that minimizes number of times of collection of a biological sample from an individual to be tested and predicts biorhythm with high precision. SOLUTION: The method for predicting biorhythm of an individual to be tested includes measuring amounts of expression of two clock genes having different phases of circadian cycle of change in the amount of expression with regard to biological samples collected from an individual to be tested three times within 24 hours and predicting the biorhythm of the individual to be tested on the basis of obtained data of time series amount of expression. The method for predicting biorhythm includes predicting biorhythm especially with high precision by collecting biological samples from an individual to be tested three times at intervals of 8 hours within 24 hours. PROBLEM TO BE SOLVED: To provide a simple and low-invasive method for obtaining information on the biological rhythm in an individual organism. SOLUTION: The method for obtaining the information on the biological rhythm in the individual organism based on the change in expression level of a clock gene in a hair follicle cell in the individual organism over time is provided. According to the method, the phase shift in the biological rhythm in the individual organism can be detected by producing and checking a molecular clock table on the individual organism two or more times. Further, the phase shift in the biological rhythm in the individual organism can be detected by checking the expression level of the clock gene at a predetermined clock time against the molecular clock table. PROBLEM TO BE SOLVED: To elucidate unexplained parts of controlling mechanism of circadian rhythm. SOLUTION: Promotion of expression inducement of Bmal1(brain-muscle ARNT-like protein 1) by RORα (retinoic acid receptor-related orphan receptor alpha) and promotion of expression inducement of Bmal1 in low oxygen state are newly found out. Existence of control mechanism of circadian rhythm that promotion of expression of RORα by the low oxygen state or the like promotes expression inducement of Bmal1 and the promotion of expression inducement of Bmal1 promotes bonding of Bmal1 with CLOCK and promotes expression inducement of Per gene or Cry gene, is much suggested thereby.
