Abstract:
PROBLEM TO BE SOLVED: To obtain a protein which can neutralize the action of TNFα (tumor necrusis factor α). SOLUTION: This protein has molecular weight of about 42,000 daltons measured by SDS gel electrophoresis and has at the N-terminus an amino acid sequence; Xaa Thr Pro Tyr Ala Pro Glu Pro Gly Ser Thr Cys Arg Leu Arg Glu (Xaa is H, phenylalanine residue (Phe) or an amino acid sequence; Ala Phe, Val Ala Phe, Gln Val Ala Phe, Ala Gln Val Ala Phe, Pro Ala Gln Val Ala Phe or Leu Pro Ala Gln Val Ala Phe), and a mutein is obtained by subjecting the protein having the TNFα suppressing action to a proper substitution, deletion or addition of an amino acid or peptide or the change of a glycoside group without strongly lowering the TNFα suppressing action.
