PURPOSE: A method for enhancing target protein accumulation by suppressing storage protein expression of a plant is provided to effectively suppress glutelin gene group. CONSTITUTION: A polynucleotide having RNAi in a plant contains a nucleotide sequence of sequence number 1. The glutelin expression is suppressed by introducing a recombinant expression vector having the polynucleotide with nucleotide of sequence number 1 and a promoter to a plant. A method for enhancing the target protein accumulation comprises: a step of introducing the recombinant expression vector to the plant; and a step of introducing the target gene to the plant.

PURPOSE: A plant transformation method using a carrier and a transformant are provided to be widely used as a core technique in biological technology. CONSTITUTION: A carrier for transformation contains at least two restriction enzyme cleavage sites which are cleaved to protrude at least one T base at 3' in order to insert a PCR product. The carrier contains at least two restriction enzyme cleavage sites which are cleaved to protrude a T base at 3' in order to insert a PCT product between a promoter and a terminator. The restriction enzyme is AhdI, Eam1105, BciVI, BmrI, HphI, Hpy188I, HpyCH4III, or MboII. The cleavage site is 5'-CTTGG-3' or 5'-CCTGG-3'. A transformant is prepared by transforming a host cell with the carrier.

The present invention relates to a BrSTY1 promoter consisting of a nucleotide sequence of SEQ ID NO: 1 and specifically inducing expression in stem apical meristem, leaves, flowers, and pods, and more specifically, to a promoter inducing an expression specifically in stem apical meristem, leaves, flowers, and pods, a recombinant expression vector comprising the same, and a plant transformed with the recombinant expression vector. A BrSTY1 promoter of the present invention induces expression specifically in stem apical meristem, leaves, flowers, and pods thereby specifically expressing a target protein by using the promoter to improve traits of crops.

본 발명은 식물의 특정한 생장 발달 시기에 특정한 조직에서 유전자의 발현을 특이적으로 유도할 수 있는 신규한 프로모터에 관한 것이다. 보다 구체적으로, 본 발명은 식물의 개화 및 종자 성숙 시기에 꽃 조직인 화탁과 꽃잎, 꽃받침, 꽃실의 기저부 등에서 유전자의 발현을 특이적으로 유도할 수 있는 신규한 BrCYS7 프로모터에 관한 것이다. 본 발명에 따르면 식물의 개화 및 종자 성숙 시기에 꽃 조직인 화탁과 꽃잎, 꽃받침, 꽃실의 기저부, 암술대 및 꼬투리의 말단에서 유전자의 발현을 특이적으로 유도할 수 있는 형질전환 식물을 제조할 수 있다. 따라서 식물의 생장 발달 단계 중 상기 특정 조건 하에서 유전자 생성물의 효율적인 생산을 위해 유용하게 활용될 수 있다.
