Abstract: The present disclosure is directed to surface modified materials such as stationary phase materials for performing size exclusion chromatography. Aspects of the present disclosure feature materials surface modified with a moiety including a polyethylene glycol (PEG) functionality and a moiety comprising a diol functionality, each of those moieties at a coverage density of at least 0.05 μmol/m2. Such surface modified materials exhibit a reduced propensity for ionic and hydrophobic secondary interactions.

Abstract: The exemplary embodiments may provide liquid chromatography systems that are smaller in size and with reduced extra-column volume than conventional liquid chromatography systems. The exemplary embodiments may reduce the size of the liquid chromatography systems enough that the liquid chromatography systems of the exemplary embodiments may be deployed adjacent to automated sample preparation robotics, adjacent to a process stream, or adjacent to the inlet of a mass spectrometer. In addition, the exemplary embodiments may reduce the extra-column volume of the liquid chromatography system by eliminating many of the connection tubes found in conventional liquid chromatography systems and by situating components of the liquid chromatography system in closer proximity.

Abstract: In some aspects, the present disclosure pertains to sample treatment methods that comprise: contacting an acidic elution solution that is free of primary amine, secondary amine and thiol groups with a sorbent having bound target antibody and separating the elution solution from the sorbent, thereby releasing bound target antibody from the sorbent and forming a first collection fraction that comprises the elution solution and released target antibody; contacting the sorbent with a neutralization buffer solution that is free of primary amine, secondary amine and thiol groups and separating the neutralization buffer solution from the sorbent, thereby forming a second collection fraction that comprises the neutralization buffer solution; and forming a neutralized solution that comprises the first collection fraction and the second collection fraction. In other aspects, the present disclosure pertains to kits for performing such sample treatment methods.

Abstract: Exemplary embodiments integrate a tee or valve into an outlet end fitting of a liquid chromatography column. The tee or valve is suitable for providing additional fluidic flow paths to ports of the end fitting and eliminates the need for post-column fluidic conduits connecting to tees or valves to insert fluidic inputs or divert flow to outputs. This integration decreases the distance that eluent from the liquid chromatography column has to travel to reach a detector relative to systems that use external tees or valves while providing tee/valve functionality and reducing the fluidic volume post-column. As a result, the exemplary embodiments help decrease sample dispersion.

Abstract: Described is a mixer 50 for a liquid chromatography system. The mixer includes a flow distributor 52, a mixing disk 54 and a flow collector 56. The mixer includes an inlet face, an outlet face and a plurality of channels each having an inlet end at the inlet face and an outlet end at the outlet face. The channels have a flow direction anisotropy between the inlet and outlet faces. A compositional solvent stream received at the flow distributor 52 is distributed across the inlet face of the mixing disk 54 and the flow collector 56 collects the distributed solvent composition stream after passing through the mixing disk 54. The mixing disk 54 may include a dispersive medium having a random porous structure. A retention time distribution of the mixer may depend on the structure of the channels between the inlet and outlet faces of the mixing disk.

Abstract: The present disclosure discusses a method of separating a sample (e.g., pharmaceutical drug, genotoxic impurity, biomarker, and/or biological metabolite) including coating a metallic flow path of a chromatographic system; injecting the sample into the chromatographic system; flowing the sample through the chromatographic system; separating the sample; and analyzing the separated sample using mass spectroscopy. In some examples, the coating applied to the surfaces defining the flow path is non-binding with respect to the sample – and the separated sample. Consequently, the sample does not bind to the low-binding surface of the coating of the flow path. The applied coating can increase the chromatographic peak area for the sample of the chromatographic system.
