    Abstract: PURPOSE: A method for producing cloned canidae is provided to conditionally express a specific gene using a Tet-on system. CONSTITUTION: A method for producing cloned canidae is as follows. A donor nucleus cell is prepared from a somatic cell collected from a canine animal. A DNA structure containing a target gene and rtTA2S-M2 trans activator sequence is introduced into the donor nucleus cell and is expressed. The donor nucleus cell with the target gene and rtTA2 S-M2 trans activator sequence is cultivated. The cultivated donor nucleus cell is transplanted to a recipient to prepare a nuclear transfer embryo and the nuclear transfer embryo is activated. The activated nuclear transfer embryo is transplanted to a surrogate mother.

CONSTITUTION: A method for producing a cloned dog by nuclear transplantation of somatic cells or stem cells comprises a step of transplanting nucleus donor cells to a denucleated oocyte of late meiosis stage II. The nucleus donor cell is somatic cells or stem cells of mitosis G2/M phase. The stem cell is derived from an embryo or adult. The somatic cells are fibroblasts or cumulus cells.
