Abstract: (I) has one or two extra amino acids substd. or inserted in the 277-527th aminoacid region. Also claimed is a DNA sequence that codes for (I) and vectors contg. the appropriate gene sequence. The DNA sequence can be cloned from human uterine tissue, by isolating mRNA and transcribing into double-stranded cDNA. After inserting the cDNA into the conventional cloning vector pUC9, a cDNA library is built up. The cDNA clone can be isolated. USE/ADVANTAGE - In the treatment of thrombolysis, (I) has an improved clotting specificity, longer half life, lower inhiBitOr binding and/or h¦gher proteolytic activity than unmodified human tissue plasminogen activator.
