Abstract: PROBLEM TO BE SOLVED: To measure fluorescence, luminescence and an absorption characteristic of a sample at a plurality of positions in a single sample well of a microplate, by setting an arrangement tool for moving a relative position of a sample-fixing tool and a detector. SOLUTION: A radiation from a light source 105 illuminates a sample 101 through an adjustment section 107 and an optical component 109. A fluorescence and/or a luminescence from the sample 101 forms an image on a detector 111 through an optical component 113 and an adjustment section 115. A second detector 117 is arranged below the sample 101 and used for the measurement of absorption. A fixing tool 103 is connected to a pair of arrangement tools 123 and the arrangement tools 123 allow the sample 101 to move within two orthogonal directions (X, Y) to the light source 105 and detectors 111, 117. A data processor 125 controls the system and is used to store data output from the detectors 111, 117.

Abstract: A method and apparatus of analyzing samples contained in a microplate is provided. The instrument is capable of measuring fluorescence, luminescence, and/or absorption within multiple locations within a sample well. The instrument is tunable over the excitation and/or detection wavelengths. Neutral density filters are used to extend the sensitivity range of the absorption measuring aspect of the instrument. Due to the wavelength tuning capabilities of the instrument, the spectral dependence of the measured fluorescence, luminescence, and absorption of the materials in question can be analyzed. The combination of a data processor and a look-up table improve the ease of operation of the instrument. Several different formats are available for the output data including creation of a bit map of the sample.

As the reaction is occurring, the substrate is exposed to a phosphor screen which absorbs the chemiluminescent emission in an image of the pattern formed by the macromolecules. The absorbed energy is then released and read, providing qualitative and optionally quantitative information regarding the immobilized macromolecules.

Distinct labels, which react with a mixture of chemiluminescent substrates to emit light at correspondingly distinct wavelengths, may be used with a phosphorescent screen selectively excitable by light of said distinct wavelengths. By this method different macromolecules can be detected simultaneously.
