Abstract:
Arrays of integrated analytical devices and their methods for production are provided. The arrays are useful in the analysis of highly multiplexed optical reactions in large numbers at high densities, including biochemical reactions, such as nucleic acid sequencing reactions. The devices allow the highly sensitive discrimination of optical signals using features such as spectra, amplitude, and time resolution, or combinations thereof. The devices include an integrated diffractive beam shaping element that provides for the spatial separation of light emitted from the optical reactions.
Abstract:
An optical instrument monitors PCR replication of DNA in a reaction apparatus having a temperature cycled block with vials of reaction ingredients including dye that fluoresces in presence of double-stranded DNA. A beam splitter passes an excitation beam to the vials to fluoresce the dye. An emission beam from the dye is passed by the beam splitter to a CCD detector from which a processor computes DNA concentration. A reference strip with a plurality of reference emitters emit reference beams of different intensity, from which the processor selects an optimum emitter for compensating for drift. Exposure time is automatically adjusted for keeping within optimum dynamic ranges of the CCD and processor. A module of the beam splitter and associated optical filters is associated with selected dye, and is replaceable for different dyes.

Abstract:
A portable spectrometer device includes an illumination source for directing at a sample, and a tapered light pipe (TLP) for capturing light interacting with the sample at a first focal ratio and for delivering the light at a second focal ratio lower than the first focal ratio. A linearly variable filter (LVF) separates the captured light into a spectrum of constituent wavelength signals; and a detector array, including a plurality of pixels, each of the plurality of pixels disposed to receive at least a portion of a plurality of the constituent wavelength signals provides a power reading for each constituent wavelength. Preferably, the TLP is lensed at one end, and recessed in a protective boot with stepped inner walls. The gap between the TLP and LVF is minimized to further enhance resolution and robustness.

The main body portion includes an upper portion that defines an upper chamber and a lower portion that defines a fluid chamber. The filter assembly is movable along a filter assembly path between a first position and a second position. The filter assembly has an opening defined therethrough. In the first position, the opening partially defines the upper chamber and in the second position the opening partially defines the fluid chamber.
Abstract:
Fluorescence chromophores such as phenylene diamine derivatives can undergo oxidative coupling and polymerization to form optical, colorimetric and fluorogenic, multimers and polymers. The presence of carbonyl containing moieties such as aldehydes and ketones under favorable environmental conditions can initiate, catalyze, accelerate and modulate this reaction which in turn provides a mechanism for the detection and quantitation of such moieties. Selected phenylene diamine derivatives can be used for the detection and quantitation of aldehyde and ketones via measurement of the reaction and the associated spectroscopic transformation. In particular, the use of meta-phenylene diamine (mPDA) and related compounds for aldehyde detection and quantitation is described. The method provides a convenient means for monitoring aldehyde and ketone levels without use of separation steps. The method is applicable to kinetic and quasi-endpoint detection assay formats.
