PURPOSE: A microorganism E. coli SB012 (KFCC 11203) sensitive to organic solvents and a method of preparation thereof are provided, thereby detecting toxicity more sensitively. CONSTITUTION: A microorganism E. coli SB012 (KFCC 11203) having improved sensitivity to organic solvents is prepared by modification or deletion of lipopolysaccharide in an extracellular membrane of E. coli SB01, wherein the modification or deletion of lipopolysaccharide in the extracellular membrane of E. coli SB01 is caused by treatment of E. coli SB01 with ethyl-methane-sulfonate(EMS), and repeatedly culturing the EMS-treated E. coli SB01 in a medium containing an antimicrobial agent and an organic solvent, wherein the antimicrobial agent is tetracycline and the organic solvent is para-xylene or bile acid salts.

PURPOSE: Provided is an effective resolution method of chiral compounds containing hydroxy group or ester group, by using lipase produced by Burkholderia Cocovenenans SY-01I(KFCC 11111). CONSTITUTION: Chiral compounds(formula 1, 2, 3 4 and 5) containing hydroxy group or ester group are resolved by hydrolyzing and trans-esterifying using the solid lipase prepared by dialysis or lyophilizing, wherein the hydrolysis is carried out at 5-35deg.C with by phosphate buffer solution of pH7.09; and the trans-esterification is conducted at 0-55 deg.C with organic solvent and an acylating agent. The organic solvent is selected from n-hexane, ethylacetate, benzene, toluene, THF, diethylether, acetonitrile, dichloroethane, dichloromethane, 1,4-dioxane, methanol, DMSO, DMF and hexane/ethylacetate(9/1 -5/5) and the above acylating agent is chosen from vinylacetate, isopropenylacetate, anhydrous acetic acid, anhydrous propionic acid and anhydrous butyric acid.

본발명은 신균주 악시네토박터( Acinetobacter sp. ) SY-01 (상기 균주는 벌콜데리아 코코베네난스( Burkholderia cocovenenans ) SY-01로 기탁되었으나, 2002년 5월 1일자로 제출한 과학적 성질 및 분류학상의 위치 표시 등의 기록서를 통하여 분류학상의 위치가 Acinetobacter sp. SY-01로 수정되었음)과 이로부터 생산되는 입체선택적 기질특이성을 갖는 리파제에 관한 것으로, 더욱 상세하게는 항진균제의 원료로 사용되는 이트라코나졸의 중간체로서 라세믹 혼합물인 시스-2-(브로모메틸)-2-(2,4-디클로로 페닐)-1,3-디옥소란-4-메틸 아세테이트의 에스테르결합을 입체선택적으로 분해할 수 있는 리파제에 관한 것이다.

PURPOSE: A recombinant plasmid containing luxCDABE, E. coli SB01 (KFCC 11176) transformed therewith, and the use thereof are provided, therefore the stability of the recombinant plasmid in E. coli can be increased to use it for the detection of toxicity. CONSTITUTION: The recombinant plasmid pBS1 contains a vector pKLJ12 containing luxCDABE derived from Photorhabdus luminescens. E. coli SB01 KFCC 11176 is produced by transforming E. coli DH5alpha with the recombinant plasmid pBS1. A method for detecting toxicity in a sample solution comprises culturing the transformed microorganism E. coli SB01 KFCC 11176 in the sample solution in a continuous fermenter, and analyzing the luminescence of the sample solution.

The minute pressure measuring method is comprised of (a) preparing a remote measuring apparatus(2) composing a light source(5), an optical detector(6), an electronic circuit(7) for position detection and an output displaying part(8), and transmitting the light irradiated from the light source through an optical fiber bundle(3) to a place to be measured, then receiving again the light transmitted through a glass tube(1a) with a measuring probe(4) to transmit light to the remote measuring apparatus(2) through the optical fiber bundle; (b) performing real-time tracking the change of a liquid surface level based on light transmitted to the remote measuring apparatus(2), digitalizing it to display the liquid surface level and a minute pressure on the output displaying part(8).

PURPOSE: Provided are a gene coding lipase of Acinetobacter species SY-01 with stereoselective substrate specificity and a protein expressed therefrom. CONSTITUTION: A gene is characterized by coding amino acid sequence of lipase of Acinetobacter species SY-01, represented by the SEQ ID NO:2. A transformed Bacillus subtilis 168/pSYLipSM(1) (KCTC 10267BP) is obtained by transformation with a recombinant vector plasmid pSYLipSM(1) containing the gene coding the lipase of Acinetobacter species SY-01. The lipase of Acinetobacter species SY-01 is manufactured by the steps of culturing the transformed bacillus subtilis, and isolating and purifying expressed lipase of Acinetobacter species SY-01 from the cultured cell.
