Abstract:
본 발명은 레트로바이러스 발현 시스템을 이용하여 α 1G T-형 칼슘 채널이 안정적으로 발현되는 HEK293 세포를 인간 Kir2.1 유전자로 형질전환시킨 세포주에 관한 것이다. 본 발명의 HEK293 세포주는 Kir2.1 (칼륨 내부 정류 채널 (potassium inwardly-rectifying channel))을 영구적으로 추가 발현함으로써 세포 막전압을 과분극쪽으로 더 낮추어 안정적인 막전압을 발현하고, 따라서 전기생리학적 방법 이외에도 고농도의 KCl에 의하여 α 1G 가 활성화된다.
Abstract:
PURPOSE: Provided is a microorganism strain producing thermophile lipase. The above strain is obtained from the soil samples near the industrial complex, identified as a novel lipase producing strain different from known Burkholderia cepacia and named as SY-02(Burkholderia cepacia SY-02). It shows thermo-stability and high enzyme activity at the higher temperature than the melting point of fat and is thus widely useful as a lipase producing strain in the field of industry. CONSTITUTION: The strain Burkholderia cepacis SY-02(KFCC-111210) produces thermophilic lipase which is secreted into the exterior of a cell of Burkholderia cepacia SY-02, and shows activity at 40-80 deg.C, preferably 70 deg.C.

본 발명은 효모의 한 종류인 클루이베로마이세스 마르시아누스 ( Kluyveromyces marxianus )로부터 분리, 정제하여 얻은 카르보닐 환원효소 (Carbonyl Reaductase, CR) 및 이를 분리, 정제하는 방법에 관한 것이다. 본 발명에 의하여 얻어진 환원효소에 의한 환원반응은 미생물에 의한 환원반응 및 화학적 합성방법으로 얻어진 환원효소에 의한 환원반응보다 입체선택성이 높고, 비용과 시간이적인 측면에서 우수하다.

PURPOSE: A process for preparing cis-2- (bromomethyl)-2- (2,4-dichlorophenyl)- 13-dioxolane- 4-methyl alcohol having high optical selectivity is provided. A stereo-selective enzyme maintains its maximum activity because the reaction product is rapidly removed from the reaction solution during the reaction in order not to inhibit the activity of the enzyme. CONSTITUTION: A process for preparing cis-2- (bromomethyl)-2- (2,4-dichlorophenyl)- 13-dioxolane- 4-methyl alcohol having high optical selectivity comprises hydrolysis of cis-2- (bromomethyl)-2- (2,4-dichlorophenyl)- 1,3-dioxolane- 4-methyl acetate using lipase produced from Acinetobacter sp. SY-01(KFCC-11111), and recovering an alcohol product as a reaction product from the reaction solution during the reaction, wherein the concentration of the alcohol product is 0.1 to 300 mg/l; the reaction product is recovered when the conversion rate of the hydrolysis reaction is 40 to 60%; the hydrolysis reaction is carried out under conditions of 40 to 60 deg. C, pH 7.0 to 9.0 and 100 to 300 rpm; and the reaction product is recovered by column chromatography.

PURPOSE: Provided is a gene of heat-resistant glutamate racemase which show stability against high temperatures, pH, and organic solvent and is useful in manufacturing medicines and food. CONSTITUTION: Heat resistant glutamate racemase is manufactured by the steps of: isolating gene of glutamate racemase consisting of 762 bases from which 254 amino acids are synthesized, from Aquifex pyrophilus; transforming E.coli is with an expression vector pGR1 that contains the gene; culturing E.coli transformant and destroying transformant cell; and recovering heat resistant glutamate racemase from cell lysat by chromatography. The base sequence of glutamate racemase shows 26.6%, 36.1%, and 35.5% of similarity with E. coli, Bacillus subtilus, and Lactobacillus, respectively.

PURPOSE: A complex of hepatitis B virus polymerase(HBVpol) and protein p11 and a method for controlling movement of the complex in HepG2 cell are provided, thereby detecting a position of the complex in human liver cell and disclosing the carcinogenesis mechanism of liver cancer. CONSTITUTION: A complex of hepatitis B virus polymerase(HBVpol) and a calcium binding protein p11 by noncovalently binding moves to promyelocytic leukemia nuclear body(PMLNB). A method for controlling movement of the complex in HepG2 cell comprises adding a calcium ion concentration regulating agent to the HepG2 cell to increase or decrease the calcium ion concentration in the HepG2 cell.

본발명은 신균주 악시네토박터( Acinetobacter sp. ) SY-01 (상기 균주는 벌콜데리아 코코베네난스( Burkholderia cocovenenans ) SY-01로 기탁되었으나, 2002년 5월 1일자로 제출한 과학적 성질 및 분류학상의 위치 표시 등의 기록서를 통하여 분류학상의 위치가 Acinetobacter sp. SY-01로 수정되었음)과 이로부터 생산되는 입체선택적 기질특이성을 갖는 리파제에 관한 것으로, 더욱 상세하게는 항진균제의 원료로 사용되는 이트라코나졸의 중간체로서 라세믹 혼합물인 시스-2-(브로모메틸)-2-(2,4-디클로로 페닐)-1,3-디옥소란-4-메틸 아세테이트의 에스테르결합을 입체선택적으로 분해할 수 있는 리파제에 관한 것이다.
