Abstract:
PURPOSE: A medium for producing capsular polysaccharide is provided to optimize medium ingredient and culture condition and to enhance productivity of the polysaccharide. CONSTITUTION: A medium which produces capsular polysaccharide from streptococcus sp. contains 30g/l of carbon source selected from glucose, fructose, galactose and lactose, 5-30 g/l of yeast extract, and 2-10g/l of sodium chloride. A method for purifying capsular polysaccharide comprises: a step of adding sodium deoxycholate to culture medium of Streptococcus sp. to perform cell lysis; a step of adding chloroform-butanol solution to the cell lysate to obtain supernatant; and a step of adding ethanol to the supernatant to precipitate capsular polysaccharide.

본 발명은 협막다당(capsular polysaccharide)을 생산하는 연쇄상구균( Streptococcus sp.), 특히 폐렴구균( Streptococcus pneumoniae )으로부터 협막다당을 생산하기 위한 배지, 폐렴구균으로부터 협막다당을 생산하는 방법 및 이를 정제하는 방법에 관한 것으로서, 협막다당의 생산성을 증가시키고, 생산된 협막다당의 분리 정제를 간소화하여 시간을 단축시킴으로써, 협막다당을 고수율로 경제적으로 생산할 수 있다. 본 발명에 따라 생산된 협막다당은 백신제조를 위한 원료로 직접 또는 캐리어(carrier)로 사용될 수 있다. 협막다당, capsular polysaccharide, 폐렴구균, 합성배지, 포도당, 효모추출물, 염화나트륨, 인산칼륨 완충용액, 경제성, 생산성, 백신

An E.coli expression vector of neuraminidase N1 of avian influenza (AI) is provided to quickly and massively produce AI virus neuraminidase N1 and select an inhibitor of AI virus neuraminidase N1 as a therapeutic agent. A vector for expressing neuraminidase N1 of avian influenza (AI) in E.coli comprises a neuraminidase N1 gene of avian influenza (AI) in multiple cloning site of pET-23d vector. A method for producing the neuraminidase N1 gene of avian influenza (AI) comprises: a step of transforming E.coli with the vector which expresses the neuraminidase N1; and a step of culturing the transformed E.coli.
