    Abstract: PURPOSE: A microorganism Klebsiella sp. producing pullulanase and a method for producing maltosyl beta-cyclodextrin using the same are provided, thereby the maltosyl beta-cyclodextrin can be effectively and cheaply produced. CONSTITUTION: The microorganism Klebsiella sp. 192 isolated from the soil can produce pullulanase which hydrolyzes the alpha-1,6-glucosidic bond of pullulan and converts into maltotriose. The method for producing maltosyl beta-cyclodextrin comprises the steps of: purifying pullulanase from Klebsiella sp. 192; reacting an enzyme solution containing the purified pullulanase, 500 to 700 mg/ml of maltose and 80 to 150 mg/ml of beta-cyclodextrin at an appropriate temperature of about 45 to 55 deg. C and pH 4.0 to 5.0; and separating the reacting materials by subjecting the solution to chromatography.

    Abstract: PURPOSE: A thermophilic cyclodextrin(CD) biosynthase and a method for producing the enzyme from Paenibacillus sp. are provided, which can biosynthesize alpha-CD, beta-CD and gamma-CD and cyclodextrin(CD) can be effectively used in medicines, cosmetics and foods. CONSTITUTION: The thermophilic cyclodextrin(CD) biosynthase is derived from a novel microorganism Paenibacillus sp. 324 strain(KCTC 0364BP), has 75,000 dalton of molecular weight, and shows its activity at pH 6.0 to 9.0 and 50 to 70 deg. C, in which cyclodextrin(CD) can seizure organic materials from starch because it has a hydrophobic region in its center. The thermophilic cyclodextrin(CD) biosynthase is produced by incubating Paenibacillus sp. 324 strain(KCTC 0364BP) in a medium containing 1 to 2% of starch, 3 to 7% of CSL and 0.5 to 1.5% of Na2CO3 at 35 to 40 deg. C.
