PURPOSE: A manufacturing method of chiral hemiester is provided to manufacture chiral hemiesters with high optical purity in very short time. CONSTITUTION: A manufacturing method of chiral hemiester comprises a step of reacting meso-cyclic acid anhride with nucleophile in organic solvent under the presence of bifunctional organic chiral catalyst in chemical formula 1 or 2. In chemical formula 1 or 2, Y and Y' are an organic group including quinuclidine moiety, and same or different each other. Z and Z' is a substituted or non-substituted quinoline group, and same or different each other. Ar is a substituted or non-substituted aryl group, or a substituted or non-substituted heteroaryl group, and n is the integer of 1-6.

PURPOSE: A method for preparing chiral amino acid ester having high stereoselectivity is provided to economically and simply produce N-acyl amino acid ester of highly optical selectivity. CONSTITUTION: A bifunctional organic chiral catalyst compound is denoted by chemical formula 1. A method for preparing the bifunctional chiral organic catalyst comprises a step of reacting amine with 3,4-alkoxy-3-cyclobutene-1,2-dione derivative. A chiral N-acyl amino acid ester is prepared from azlactone by reacting racemic azlactone, (S)-azlactone or (R)-azlactoen with nucleophile under the presence of organic solvent.

A method for preparing a penem intermediate is provided to decrease the amount of a rhodium catalyst used significantly by using a copper catalyst as an auxiliary catalyst and produce a target product very economically by increasing the efficiency of the rhodium catalyst. A method for preparing a bicyclic keto ester compound represented by a formula(1) from a diazo compound represented by a formula(2) is characterized in that a rhodium catalyst and a copper catalyst, which is copper powder, Cu(I) compound or Cu(II) compound, are simultaneously used. In the formulae(1) and (2), R is a protecting group of a carboxylic acid which is C1-4 alkenyl, or benzyl substituted with para nitro, para C1-4 alkoxy or para C1-6 alkyl; R^1 is H, optionally substituted alkyl, optionally substituted aryl, or optionally substituted heterocyclyl; R^2 is H, acyl or silyl which is tri-substituted with alkyl and aryl, provided that the compound of the formula(1) and the compound of the formula(2) are existent in various optical isomers due to the presence of non-symmetric carbon atoms. 본 발명에 따른 키랄성 (S)-프레가발린 전구체의 제조 방법은 이작용성 키랄 촉매의 존재 하에, 베타-이소부틸 나이트로 알킨과 말로네이트를 비대칭 마이클 반응시키는 단계를 포함한다. 이에 따라 사용하기에 안전한 촉매를 이용하여 저렴하고, 인체에 유해하지 않으며 환경오염을 유발하지 않는 조건에서 높은 광학 선택성과 반응성으로 키랄성 (S)-프레가발린 전구체를 합성할 수 있다.

본 발명은 에틸렌 글라이콜이 유도체화된 화학식 1 또는 화학식 2의 촉매의 존재 하에서 시아나이드 공급원을 사용하여 스트레커 반응시키는 것을 특징으로 하는 키랄성 α-아미노나이트릴의 제조방법에 관한 것이다. 본 발명에 따르면, 다양한 구조의 키랄성 α-아미노나이트릴을 매우 높은 입체선택성으로 합성할 수 있고, 또한 비천연 아미노산인 (R)-α-아미노산의 전구체인 (R)-형태의 α-아미노나이트릴을 높은 광학 순도로 제조할 수 있으며, 시아나이드 공급원으로서 열적 안정성 및 보관성이 뛰어나고 저렴하고 사용하기 용이한 알칼리 금속 시안화물 단독 또는 알칼리 금속 시안화물과 설피닉산 알칼리금속염 또는 설피닉산의 조합을 사용하므로, 공업화에 매우 유용한 매우 경제적이며 간편한 방법이다.
