Abstract:
본 발명은 효모로부터 인간 리포코틴-I 단백질을 효율적으로 발현 분비시키는 플라스미드 벡터, 상기 플라스미드 벡터로 형질전환된 재조합 효모, 및 상기 재조합 효모의 배양을 통한 인간 리포코틴-I 단백질의 생산 방법에 관한 것으로, 본 발명의 발현 벡터는 GAL10 포로모터, 이누리나제 분비시그날, 리포코틴-I 구조 유전자 및 리포코틴-I 터미네이터를 포함하는 것을 특징으로 한다.
Abstract:
The tocopherol is separated and purifyed by (a) adding organic solvent of n-hexane, n-heptane, acetone or methylethylketone into the soybean oil distillate and removing the crystallized sterol of the mixture by filtering, (b) methylesterifying the obtained fatty acid by reflux at 60-80 deg.C with 35 % HCl catalyst, (c) dissolving in lower alcohol, adsorbing it into alkaline anion exchange resin and purifying with lower alcohol contg. acetic acid, formic acid, oxalic acid or succinic acid, (d) recirculating the esterifyed product in a molecular distillator to obtain the final product.
Abstract:
PURPOSE: Provided is a method for manufacturing invertase and yeast extract from yeast at the same time by extracting the invertase under optimal conditions to obtain the most active invertase, concentrating through membrane process to get the concentrated invertase solution and obtaining the yeast extract by self digestion method of the residue and the filtrate. CONSTITUTION: The method comprises the steps of: extracting the invertase from the shake culture medium of yeast with papain; centrifuging or micro-filtering the extract to separate the supernatant and the yeast; obtaining the invertase concentrate by ultrafiltering; and acquiring the yeast extract by self digesting the mixture of the above centrifuged or micro-filtered yeast and the above filtrate of the ultrafiltration.
Abstract:
PURPOSE: Provided are a novel bioactive peptide containing inhibiting activity of angiotensin-I-converting enzyme(ACE) and its usage for curing hypertension. Particularly, the bioactive peptide is manufactured by separating an alpha s1 casein from cDNA library of human mammary gland and expressing it in E.coli, wherein 6 of histidines are added to C terminal in order to easily purify alpha s1 casein. CONSTITUTION: A human alpha s1 casein is expressed in E.coli and purified. And then, recombinant human alpha s1 casein is decomposed into trypsin. Thereafter, ACE inhibitory peptide is separated by using high performance liquid chromatography(HPLC). Next, an amino acid sequence of the selected peptide is analysed and a molecular weight thereof is measured. Finally, an ACE inhibiting activation of peptide is analyzed quantitatively.

본 발명은 효모로부터 인체 부갑상선 호르몬(human parathyroid hormone: hPTH)을 효율적으로 생산하는 방법에 관한 것으로 단백질 분해 효소인 효모 아스파틱 프로테아제 3 (Yeast Aspartic Protease 3: YAP3) 결손 균주를 제작하여 인체 부갑상선 호르몬의 분해를 최소화하므로써 완전한 형태의 인체 부갑상선 호르몬을 생산하는 방법을 제공한다.
