Abstract: Process for the production of a protein correlated with the diphtheric toxin which comprises culturing in a liquid nutrient medium having a concentration of iron ions of from 0.05 mu g/ml to 0.5 mu g/ml, at a temperature of from 30 DEG C. to 40 DEG C., in a neutral culturing environment and under aerobic conditions, a microorganism belonging to the Corynebacterium diphtheriae genus with two mutant phages encoding the protein correlated with the diphtheric toxin integrated in a nontandem mode in their chromosomes.

Abstract: A system of inducible positive regulation of use in the construction of expression vectors in host microorganisms for controlled production of heterologous proteins in high yields. More particularly the system comprises the vir EcoRI-XhoI regionof about 4.5 kb contained in the chromosomal DNA of Bordetella which codes for one or more regulating factors, and a terminal 5' region situated in front of the promoter of the chromosomal Bordetella genes capable of being activated or repressed in trans by the aforementioned factors. Also described are expression vectors which contain the regulating system and microorganisms transformed by the vectors.

Abstract: Cloning and sequencing of the EcoRI fragment of B. pertussis chromosomal DNA with 4696 base pairs, conta ining the genes which code for the five subunits of the pertussis toxin. A hybrid plasmid containing the DNA fragment or its further fragments and a micro-organism transformed by the hybrid plasmid and capable of expressing the cloned DNA fragment or further fragments thereof by synthesis of the pertussis toxin or one or more subunits of the pertussis toxin. The pertussis toxine or one or more subunits of the pertussis toxin so obtained are useful for the preparation of vaccines and diagnostic kits.

Abstract: The cloning and sequencing of the gene which codes for a new pilinic subunit of Bordetella pertussis are described. The aminoacid sequence of the mature subunit, deduced from its necleotide sequence, is similar but not identical to that of the known pilins 2, 3 and 6. Polypeptides having the aminoacid sequence of the mature pilinic subunit or of regions thereof are particularly useful for the development of synthetic acellular vaccines against pertussis.

New pertussis toxin (PT) mutants are described being immunologically active and having reduced or no toxicity, characterized in that at least one of the aminoacid residues Glu129, Asp11, Trp26, Arg9, Phe50, Asp1, Arg13, Tyr130, Gly86, Ile88, Tyr89, Tyr8, Gly 44, Thr53 and Gly80 of subunit's 1 aminoacid sequence is deleted and substituted by a different aminoacid residue selected in the group of natural aminoacids; Bordetella strains capable of providing and secreting said PT mutants and means and methods for obtaining them are also described. The Bordetella strains and the PT mutants produced by them are particularly suitable for the preparation of effective ceilular and acellular antipertussis vaccines. (Fig. 1)
